Manyinflorescenced Sweetvetch Root(Radix Hedysari) was a kind of Chinese herbs drugs, which is the roots of Hedysarum Polybotrys Hand.-Mazz(H.polybotrys Hand.-Mazz). It is warm in property and sweet in flavours, enters lung and spleen meridian, can invigorate qi to secure the exterior, it is diuretic and can dispel virus, discharge pus, heal sore and promote granulation. There are four kinds of radix hedysari in China for its difference in productional place, but Hedysarum Polybotrys Hand.-Mazz produced in Gansu province was sure best impotant because of its best quallities and use, which also was sure genuine medicina materials in Gansu province. It abundantly distributed in Wudu district. The study was carried out to extracting polysaccharides from H.polybotrys Hand.-Mazz, investigating the reactive oxygen species scavenging effect, anti-lipid peroxide effect in vitro and anti oxidative stress capacity in vivo. Crude water-soluble polysaccharides (CHPS) were isolated from the roots of H.polybotrys Hand.-Mazz. by hot water extraction followed by ethanolprecipitation. Its total soluble sugar content was detected colorimetrically by the phenol-sulfuric acid method. Antioxidant efficacy of CHPS were evaluated using hydroxyl radical and superoxide anion radical scavenging activities. Moreover, the protective effect of CHPS for the autoxidation of mice'liver homogenates and erythrocyte hemolysis induced by H2O2 were mearsured. Furthermore, 50 wista mice were randomly divided into 5 groups. Control group(Ⅰ) were intragastric administration with distilled water everyday. Cyclophosphamide (Cp)group(Ⅱ) were intraperitoneal injected Cp at the sixth day of experiment. CHPS low, mid and high groups(Ⅳ,Ⅴ,Ⅳ) were intragastric administrated with different dose of CHPS everyday, and Cp were intraperitoneal injected. All mice sacrificed at 7th for some index including SOD, GSH-Px and that were examined to determine antioxidant effect of CHPS in vivo.The results showed that it total carbohydrates content was determined to be 26%. The results also exhibited a significant (P<0.01) concentration (0.258mg/mL) dependent mitigation of·OH scavenging as assessed by Fenton action method. Similar results were obtained when O2-.scavenging (P<0.01,120 mg/mL) as assessed by pyrogallic acid autoxidation action method. However, the results showed that a significant concentration dependent mitigation of anti lipid oxidation as assessed by MDA spontaneous formation in liver homogenates of mice at a concentration range of 0.1252mg/mL(P<0.01), once more CHPS was increased, instead of protect action was to being accelerate lipid oxidation. Similar results was obtained when CHPS protect erythrocyte hemolysis from erythrocyte induced by H2O2 at the range of 2.520 mg/mL(P<0.01), then more CHPS, more erythrocyte hemolysis. Compared with control group(Ⅰ) , the content of MDA significantly increased, the activities of SOD and GSH-Px significantly decreased in liver and kidney homogenates of Cp group mice; while compared with Cp group, the content of MDA significantly decreased(P<0.01), the activities of SOD and GSH-Px significantly increased (P<0.01)in liver and kidney homogenates of Cp group mice.These results indicated that anti lipid oxidation of CHPS may be responsible for its oxygen free radical scavenging effect. Some dose of CHPS can lifting experiment mice antioxidant level by reactive oxygen species scavenging activities and to promote SOD, GSH-Px activities. In a word, CHPS can scavenge reactive oxygen species in vitro and antioxidant in vivio.
