Objective: To study the effect of Ulinastatin on brain weight and cerebrocellular degeneration,necrosis,apoptosis and gliocyte hyperplasia of hypoxic-ischemia encephalopathy in newborn rats.Methods: select 50 newborn Wistar rats of 7-day old to establish HIE model, and there are 30-rats were intraperitoneally injected Ulinastatin immediately and after 8 hours,16 hours, the rats are divided one,two and three therapy groups and the dose are divided 0.5万U/kg, 1.0万U/kg, 2.0万U/kg three groups, every group include 10 newborn rats, then the rats were killed after hypoxic and ischemia another 24 hours. The brain tissue were weighted and fixed, the number of apoptosis was detected after the brain tissue were stained by TUNEL; the cerebrocellular degeneration,necrosis,apoptosis and gliocyte hyperplasia of the brain tissue that were stained by HE, which were measured with quantitative pathological score; observed hippocampus brain cells by electron-microscopy, and the results were compared among HIE model group, sham operation group and three Ulinastatin-therapy groups.Result: the brain total weight and weight difference from left and right brain in HIE group and therapy groups and sham operation group, therapy groups were much higher than that of sham operation group(P <0.05或0.01), HIE model group were much higher than that of therapy groups, there were no significant different in three therapy groups(P>0.05).the number of apoptosis and the brain tissue quantitative pathological score in HIE group and therapy groups were much higher than that of sham operation group(P<0.01), that of the therapy groups were much lower than that of HIE group (P<0.01) there were no significant different in three therapy groups. (P>0.05)Conclusion: Ulinastatin can decrease cerebrocellular edema,cerebrocellular degeneration,necrosis,apoptosis. have prevention and cure effect on HIE.
