OBJECTIVE: Cisplatin-resistant human ovarian carcinoma cellline SKOV3/DDP was cultured with graded concentration of lobaplatin indifferent time, then explore the growth inhibition, inducing apoptosis andcell cycle arrest effect of lobaplatin on SKOV3/DDP cell line. To identifywhether lobaplatin can treatment the patients of cisplatin-resistantovarian carcinoma or not.METHODS:1. SKOV3/DDP cell line for cisplatin-resistant humanovarian carcinoma was cultured to logarithmic growth phase.2. Cell proliferation in different concentration and time wasdetermined by MTT (Methyl thiazolyl tetrazolium) assay. And compared withA value of cisplatin in identical time and concentration.3. The characters of apoptosis were assessed by light and transmissionelectron microscopy. Then the changes of lobaplatin and cisplatin on cellSKOV3/DDP in identical concentration were found by light microscopy.4. Flow cytometry was used to measure the induction of cell cyclearrest and apoptosis rate.RESULTS: The result of MTT colormetric assay showed thatproliferation of SKOV3/DDP cell was inhibited by lobaplatin in adose-time-dependent fashion. And there was significant (P<0.05) between the A value of lobaplatin and cisplatin. When SKOV3/BDP cell line wastreated with lobaplatin at a certain concentration, the morphologicchange of apoptosis was found by light, transmission electronmicroscope. And observed the cell population decresed obviously in cellon lobaplatin in light microscope.Flow cytometric analysis showed thatafter using lobaplatin, cell cycle distribution changed at first and asthe time prolonged apoptotic peak went higher.CONCLUSION: apoptosis induced by Lobaplatin is one of themechanism Which inhibits proliferation of SKOV3/DDP cell line. AndG0/G1 phase arrest may be related to the mechanism.
