| The diabetes (Diabetes Mellitus, called DM) is a metabolism disorder syndrome with the chronic high blood sugar connected with the heredity factor and the many kinds of environmental factor. DM is endocrine metabolism abnormality disease which takes place often and is hard to permanently cure with higher complication and disease incidence rate. DM already became the third bigest sickness only inferior after the tumorand and cardiovascular disease. By using streptozotoc (STZ) to establish Gestational Diabetes Mellitus (GDM), we studied the effect of GDM on development of young rats in different time. The GDM has huge possibilities to develop in 2 diabete and its pathogenesis is similar very much with 2 diabetes. The polysaccharide is the important active constituent of Chinese native medicine and has the effect to fall in the blood sugar. The polysaccharide is one kind of immunity conditioner, has the effect to fall in the blood fats, and showed many kinds of biological activities with non-toxic side effect also. By using STZ to establish 2 diabetes big rat model, we studied the effect of Artermisia Polysaccharides (ASP) on falling in the blood sugar and its mechanism. The purpose is trying to find the way to develop one kind of new natural medicine on the diabetes assistance treatment.1 Effect of gestational diabetes mellitus on the development of pancreas and the liver antioxidation of young ratsTo establish GDM model, the rats were intraperitoneally injected STZ at the dosage of 35mg/kg.bw one time on 1st day of the pregnancy in the experiment model 1, 7th day of the pregnancy in the experiment model 2, and 14th day of pregnancy in experiment the model 3. The livers of young rats were dissected and the superoxide dismutase (SOD) activeness and alondialdehyde (MDA) the content were determined separately on growing to 18 day, 20 day and the birth. The the pancreas organization was fixed in 4% paraformaldehyde and HE dying was used to observe histological changes. The result showed that the liver SOD activity is lower at each GDM model group than that at the normal control group in the developing period obviously, the content in liver MDA is higher in GDM model group than that in the normal control group. Therefore, the anti-oxidized level of young rats liver of GDM model group was lower. Histological observation showed that the development of pancreas in GDM model groups was in advance compared with normal contral group and the islets of pancreas in GDM model groups was bigger than normal control group. Moreover, the effect of DM at initial, intermediate, advanced stage pregnancy are smaller on the development of young rats gradually.2 Functional and mechanism study of polysaccharides from Artermisia polysaccharides (ASP) on decreasing blood sugarArtermisia Polysaccharides (ASP) was extracted by the supercritical extract, the water distill and alcohol precipitates repectively. The protein was withdrawn by the papain +Seveage law albumenose-free method. 2 diabetes model was induced by STZ. The experiments were divided into 5 groups. That is normal control group, the control group of ASP, the diabetic group (model group), Diabetes + ASP high dose group (high-dose group) and diabetic + ASP low dose group (low-dose group). ASP was given at dose of 188mg/ kg.bw at ASP control group, 250mg/kg.bw at high dose group and 125mg/kg.bw at low-dose group.ASP was fed during 36 days consecutively every day. At the 36th day, animal was killed after fasting for 12 hours, blood was taken from heart. Result showed that glucose was decreased (P<0.01), serum insulin was increased (P <0.01), serum glucagon was decreased (P <0.01), serum glutamic pyruvic transaminase, serum aspartate aminotransferase decreased (P <0.01) respectively at high-dose group compared with the model group. The normal control group islet was observed by In situ hybridization technique. The ultrastructure and HE staining showed that secretory granules ofβ-cell of islet pancreas in model group reduced and the morphous of theβ-cell was abnormal.β-cell of islet pancreas was hyperplasia, islet cell secretory granules was increased significantly, the symptoms of diabetes reduced at the high dose group compare with model group. The mechanism of ASP effect on lowed blood sugar of DM might be contribute to the restoration ofβ-cell of islet, promotion of islet insulin secretion, regulation of insulin and glucagon secretion. |
