Objective By intervention high fat and sugar diet to investigate agestational diabetes mouse model establishing method which issteady,reliable and similar to human being.Methods C57BL/6J micewere randomly divided into normal diet group and high-fat and sugarfeeding group, and then each group is divided into two subgroups, highfat and sugar diet of pregnant group and high-fat and sugar dietnon-pregnant group, normal diet of pregnant group and normal dietnon-pregnant group.From the date of pregnancy success, the normalcontrol group was given normal diet, high fat and sugar feeding groupwas given high fat and sugar feeding. Before pregnancy and pregnancy10,18days measured fasting plasma glucose and body weight, andobserve the water intake and urine output change, the third trimester ofpregnancy measuring insulin levels and insulin resistance index, themodeling rate of the gestational diabetes mouse were computed.Results In the high fat and sugar diet of pregnant group,the bloodglucose levels increased gradually over pregnancy,and were morestable.The body weight of the high fat and sugar feeding group was also higher than that in the normal diet group. The insulin levels and insulinresistance index were increased in the third trimester of pregnancy.Themolded rate of gestational diabetes is53.3%.Conclusion The high fatand sugar feeding C57BL/6J pregnant mouse to establish a mouse modelof gestational diabetes has good stability, and are similar to humanbeing of gestational diabetes. Objective: The aim of this study to measuring the blood cholesteroland triglyceride levels of the high fat and sugar diet-induced mousemodel of gestational diabetes exposed of the characteristics of lipidmetabolic. Methods: After the end of the first part of the experiment,the serum blood cholesterol and triglyceride content was measuredusing enzyme immunoassay. Observed the pathological changes of livertissue staining HE. Results: The cholesterol and triglycerides of thehigh fat and sugar pregnant group were higher than the high fat andsugar nonpregnant group, the normal diet nonpregnant group and thenormal diet pregnant group.Liver pathology shows that in the high fatand sugar pregnant group the mouse hepatic board arrangedirregularly,there are fat vacuoles of varying sizes in the cytoplasm ofliver cells. Conclusion: The GDM mouse model induced by high fatand sugar diet with abnormal glucose metabolism has abnormal lipidmetabolism, and the pathological changes of the liver tissue.They aresimilar to lipid metabolism of human GDM.
