Objective: to establish the specific immunological indexes of sensitized dendritic cellby observing the change of IFN-γ,IL-10,IL-12 in the peripheal blood irgastrointestinal cancer before and after dendritic cell combined withchemotherapy, and observe the clinical curative effect and side effect of thecombined chemotherapy in gastrointestinal cancer.Methods: forty patients with advanced gastric cancer or colorectal cancer wererandomly divided into chemotherapy group (control goup) and dendritic cell combinedwith chemotherapy group (observing group), sensitized matured DCs were derived fromperipheal blood monocytes of the patients in observing group before chemotherapywhich were loaded tumor lysate and cultivated in vitro, in the course of cultivation, the patients in observing group received oxaliplatin (L-oHP) plus 5-FU/CF, sensitizedmatured DCs were administered after chemotherapy, the patients in control groupreceived the treatment of the same regimen, the concentrations of IFN-γ, IL-10, IL-12 in blood serum were detected before and after treatment; toxicity, clinical benefit response rate and KPS were obseved.Results: the concentrations ofIFN-γ, IL-12, IL-10 in blood serum and KPS werenot significant difference in two groups before treatment. In observing group, theconcentrations ofIFN-γ, IL-12 in blood serum and KPS were significantlyincreased, and the concentration of IL-10 was decreased afte treatment, in contolgroup, the concentrations of IFN-γ, IL-12 in blood serum and KPS weredecreased, and the concentration of IL-10 was increased after chemotherapy, there was statistical difference. No statistical difference was found in clinical benefit responserate between two groups(P>0.05).Compared with control group, bone marrowsuppression was mild in obversing goup(P<0.05). In observing group, when sensitizedmatured DCs were administered, fever was a common side effect, serious side effectwas not obversed.Conclusion: IFN-γ,IL-10, IL-12 are the specific immunological indexes ofsensitized dendritic cell therapy; The response rate of biochmotherapy is the samewith chemotherapy; dendritic cell can decrease side effect of chemotherapy, changeimmunosuppression, enhance immune function and improve patients' quality of life.
