Purpose To observe the histo-morphological heterogeneity in progression, investigate the expression ofβ-catenin,MMP7, cyclin D1 and c-myc in normal brain and glioma, and evaluate the relation between histo-morphological heterogeneity and the expression ofβ-catenin,cyclin D1,c-myc and MMP7 in various grade of glioma.Methods The sections of 89 glioma stained with hematoxylin and eosin were observed. tissue microarray were constructed to contain 89 glioma and 12 normal brain tissues. S-P immunohistochemical staining was used to examine the expression ofβ-catenin,cyclin D1,c-myc and MMP7 in various grade of glioma on the tissue array sections.Results 89 glioma were divided into three types: astrocytes, oligodendrocytes and ependymal cells and 11 variants according to the histological differentiation. These gliomas were composed of 47 low grade (Ⅰ-Ⅱgrade) and 42 high grade (Ⅲ-Ⅳgrade) according to the criteria of WHO(2000).Positive rate ofβ-catenin expression in low-grade (Ⅰ-Ⅱgrade) glioma was 53.2%(25/47) with cytoplasmic staining predominantly and faint expression in nuclear, compared with a higher positive rate of 88.1%(37/42) in high-grade glioma(Ⅲ-Ⅳgrade),which showed nuclear expression in 80%(34/42) cases and more intense staining. In low-grade and high-grade glioma the positive rates of cyclin D1,c-myc and MMP7 were 63.8 %(30/47):83.3%(35/42),70%(33/47):95%(40/42) and 68.1%(32/47):88.1% (37/42) respectively. No positive reaction was observed in control group. The expression ofβ-catenin,cyclin D1,c-myc and MMP7 were significantly different in low-grade and high-grade glioma(p<0.05, p<0.05, P<0.01 ,P<0.05 respectively).The expression ofβ-catenin had close correlation to that of cyclin D1,c-myc and MMP7 in glioma (k=0.808,P<0.000; k=0.699,P<0.000; k=0.376,P<0.000 respectively).Conclusions (1) The histo-morphological differentiation and the grading of glioma reflect the heterogeneity of glioma. (2)The accumulation of 13 -catenin in cell plays a role in the progress of glioma, might have correlation with pathogenesis, development and invasion ,and can reflect the progress of heterogeneity such as malignancy and pathological grading. (3)β-catenin and its down streamtarget genes cyclinD1,c-myc and MMP-7 are all play roles in progress of glioma, and promote the pathogenesis, development and invasion of glioma.(4)β-catenin can regulate glioma by the change of cyclinD1,c-myc and MMP-7,as the expression of cyclinD1,c-myc and MMP-7 increase with abnormal increasing ofβ-catenin.
