Objective To explore the expression and significance of early growth response-1(Egr-1) in different stage of selenite-induced cataract and in lens epithelial cells which are impaired by H2O2.Methods 1.45 Sprague-Dawley rat pups was divided into control group,normal group and experiment group. The experiment group was subcutaneous injected of 3.46mg sodium selenite/kg body weight, while the control group was subcutaneous injected of corresponding sodium chloride, nothing was injected to the normal group. After injecting sodium to Sprague-Dawley rats,the degree of lens opacity was observed by slit-lamp. On d1,3,7, the Egr-1 expression was detected by immunohistorychemistry(SP assay) respectively.2. The epithelial cells was cultured in vitro,after treated by H2O2, the Egr-1 expression was detected by immunohistorychemistry(SP assay).Results 1. In the experiment group,the lens opacity continuously aggravated, dust opacity and crico-opacity were formed d 3 after injecting and typical nuclear cataract was formed at d 7 after injecting. While in the control and normal group,the lens were keeping transparent2. The Egr-1 expression was weakly positive at d 1 after injecting,positive or hadro-positive at d 3 after injecting, while weakly-positive at d 7 after injecting .While in the control and normal group, and Egr-1 expression was negative.3. 2h after treated with H2O2 ,the expression of Egr-1 in the epithelial cells was hadro-positive ;the opatical density value was 0.55±0.08;8h after treated withH2O2, the expression of Egr-1 in the epithelial cells was positive ;the opatical density value was 0.40±0.09; The difference between the two groups was significant ,while 24h after treated with H2O2 and in control group the expression of Egr-1 in the lens epithelial cells was negative.Conclusion Egr-1 was expressed at early stage of selenite-induced cataract, and it also expressed in the early stage of the epithelial cells afer treated with H2O2, it may be one of the early effective factors in the development of oxidative damaged cataract.
