| Background and Objective: FGA-128C/G, as one of gene polymorphism sites of alpha-fibrinogen, was identified in recent years. This polymorphism site locates in the promote region of the fibrinogen alpha chain mRNA transcription, and this polymorphism variant may change the promote region. Plasma fibrinogen levels and fibrinogen may be influenced by this genetic modification, in addition. But no association on FGA- 128C/G in plasma fibrinogen levels and the risk of cerebrovascular disease have been reported in China. The -128C/G restriction fragment length polymorphism of the FGA gene was used in a case-control study is used to investigate the genetic variation at this locus in relation to plasma fibrinogen concentrations and the risk of cerebral infarction. The aim of this study is to demonstrate the molecular machnism of cerebral infarction (CI) and provide theoretical evidence for the prevention and treatment of CI.Methods: 194 patients suffered from CI within 7 days after the event and 60 healthy control individuals from Xiangya Hospital of Zhongnan University were enrolled in this trial.The FGA-128C/G polymorphism was analyzed by PCR-RFLP, and plasma fibrinogen levels were obtained from Clauss assay. Statistical analysis was carried out by SPSS11.5 software.Results : The frequencies of CG, CC genotypes were 14.3% ,85.7%, respectively and that no higher in patients with cerebral infarction(CI) than in control subjects(p>0.05). FGA-128C/G polymorphism is not associated with plasma fibrinogen levels. It seems that there is no association between plasma fibrinogen level and the genotypes (CG, CC).The GG genotype was not found in this study. The frequencies of C and G alleles were no higher in patients with cerebral infarction. There was association between the plasma fibrinogen level and the increasing of age (regression coefficient, 0.079). There was difference about the plasma fibrinogen level and sexes, also (regression coefficient,-0.534) .Conclusion: There are FGA- 128C/G polymorphisms in Chinese Hunan Han ethnic population. The FGA-128C/G polymorphism of fibrinogen may not associate with plasma fibrinogen levels and the risk of cerebral infarction. The plasma fibrinogen level increased according to ascending of age and the fibrinogen level of the male is higher than finale groups. |
