| Objective: To study the effect of propylthiouracil on experimental autoimmune thyroiditis(EAT), and hope to provide a new method for curing Hashimoto thyroiditis.Methods: 72 KM mice were randomly divided into 3 groups, each has 24 mice. Among of them, per group was divided into 2 subsets,each has 12 mice. A: Normal control group,divided to A1,A2;B: EAT control group and to B1,B2; C: propylthiouracil treatment group divided to C1,C2; B, C groups were immunized with thyroglobulin(Tg) to establish the model of EAT and were administered from ten days after primary immunity. B group was given distilled water, B1 group was observed for 4 weeks and B2 group for 6 weeks. C group was administered Propylthiouracil(20mg/d),C1 group was observed 4 weeks, and C2 group was observed 6 weeks. A group was given distilled water as control groups, A1 group was observed 4 weeks, and A2 group was observed 6 weeks. A1,B1,C1 group were all killed after given drugs for 4 weeks,and A2,B2,C2 group were killed after given drugs for 6 weeks. We used radio-immunoassay(RIA) to test the levels of serum thyroglobulin antibody(TgAb) and thyroid peroxidase antibody(TPOAb). At the same time separate thyroid glands to histological analysis.Results: (1)EAT mice had inflammation cell infiltration in thyroid follicle matrix. The levels of serum TGAb and TPOAb were increased. (2)The incidence and the level of TGAb,TPOAb of C1 group both decreased compared with B1 group after administering 4 weeks (P< 0.05) .(3) The incidence and the level of TGAb,TPOAb of C2 group both decreased compared with B2 group after administering 6 weeks (P<0.05). (4)The incidence and the level of TGAb,TPOAb of C2 group both decreased compared with C1 group (P<0.05) .Conclusions: Propylthiouracil has some effects on EAT and the curative effect was enhanced along with extending the time of therapy. |
