| Objective:To explore the role of adiponectin and TNF-αin the development of diabetic retinopathy(DR)in diabetes mellitus rats.And to provide a new method to diagnose early and therapy DR effectively.Methods:64 Wistar rats were divided randomly into two groups.32 rats in normal control group and 32 rats in diabetes mellitus group.The subjects in diabetes mellitus group were injected intraperitoneally with dose STZ 50mg/kg while the ones in normal control group were injected intraperitoneally with the similar dose of citric acid buffer.At 1,4,8 and 12 weeks, weight,fast Blood glucose,serum adiponectin,TNF-α,C-peptide were measured,the pathological changes of retina were examined using light microscope and electron microscope,retinal expression of adiponectin and TNF-αwere determined by immunohistoehemistry.Meanwhile,the changes of retinal capillaries basement membrane and immunohistoehemistry were measured by analyse morphologically.We compare these data with analysis of variance,and analyse the correlation of adiponectin and TNF-α.Results:There were a significant difference in the levels of fast blood glucose(FBG) between normal control group and DM group at every stages(P<0.01),the levels of FBG of the DM group were higher than that of the control group.Compared with the control group,there were significant decrease in the level of C-peptide,in the DM group.With the development of DM,membrane disks spaces in rods of DM were enlarged gradually,dividing and dissolving locally.The capillaries basement membrane increased gradually,and capillaries became narrow and even occluded.The mitochondria swelled and even denatured.The increase of serum adiponectin were observed at 4,8 and 12 weeks and serum TNF-αduring the whole experiment;immunohistochemisty showed that in DM group the expression of TNF-αwas increased continually during the whole experiment(1 week P<0.05,4,8 and 12 weeks P<0.01) and the expression of adiponectin was increased at 4,8 and 12 weeks.There was a significant positive correlation between the expression of TNF-αand adiponectin in retinal tissue of DM group(r=0.872,P<0.01).Conclusion:The increase of adiponectin and TNF-αin serum level and the expression in retinal tissue may play a important role in the pathogenesy of DR.adiponectin may play a protective role,and meanwhile TNF-αis harmful in the progression of DR. |
