A Preliminary Study On The Relationship Between Adopnnectin,TNF-α And Diabetic Nephropathy In DM Rats

Objective:To explore the role of Adiponectin(ADPN)and TNF-αin the development of diabetic nephropathy(DN)in diabetes mellitus rats.And to provide a new method to diagnose early and therapy DN more effectively.Methods:64 Wister rats were divided randomly into two groups.32 rats were in normal control groups and the others were in diabetes mellitus groups.The subjects in diabetes mellitus groups were injected intraperitoneally with dose STZ 50mg/kg while the ones in normal control group only done with the similar dose of citric acid buffer.At 1,4,8 and 12 weeks,weight,fast Blood glucose and 24-hour urinary albumin excretion were observed,serum ADPN,TNF-α,fast blood C-tide,BUN and Cr were measured,the pathological changes of kidney were examined by light microscope and electron microscope.Renal expression of ADPN and TNF-αwere determined by immunohistoehemistry.Meanwhile,the changes of glomerular volume,GBM and immunohistoehemistry were measured by analyzer morphologically.We compare these data with analysis of variance,and analyze the correlation of ADPN and TNF-α.Results:Compared with the control group,diabetic rats expressed more urinary albumin excretion,12week later,it decreased but was still higher than the control group;path histological study revealed that there was more glomerular matrix and thicker GBM in diabetic rats (P＜0.01),the volumes of glomerule of diabetes mellitus groups were higher than the control (P＜0.01);the increase of serum ADPN and TNF-αwere observed during the whole experiment(P＜0.01);immunohistochemisty showed that the expression of ADPN and TNF-αin renal tissue in diabetes mellitus groups were much higher than the normal control groups (P＜0.01),then they increased continually during the whole experiment.There was a significant positive correlation between the expression of TNF-αand ADPN in renal tissue of DM group(r =0.761,P＜0.0001).Conclusion:The increased of ADPN and TNF-αin serum level and the increased expression of them in renal tissue may play a important role in the pathogens of DN.TNF-αmaybe advance the progression of DN,ADPN may also have the potency to enhance the developing of this disease.
