| Objective To establish impaired glucose tolerance(IGT)animal model.To explore the role of adiponectin and liver in IGT rats.Methods 60 Wistar male rats were randomly distributed into 2 groups:control group and high fat-fed group.Setting 2 stages as 8,12 week during evolution of IGT.We recorded the changes of body weight every week and did oral glucose tolerance test(OGTT)every two week, measured blood fat series,insulin and adiponectin at 8,12 week,assessed the degree of insulin resistance by using homeostasis model assessment of insulin resistance(HOMA-IR).The hepatic pathological changes were observed using light microscope.Adiponectin receptor 1(AR1) mRNA and protein were respectively examined by reverse transcription-polymerase chain raction(RT-PCR)and western blotting.Results①9 rats' 2h blood glucose achieved the diagnostic criteria of impaired glucose tolerance in high fat-fed group at 12 week,while its fasting blood glucose increased than control group(P<0.05)but not achieved the diagnostic criteria of impaired fasting blood glucose;②In high fat-fed group weight,visceral fat content/weight ratio(VF/W),insulin and HOMA-IR increased obviously than that in control group(P<0.05)at 8,12 week,as well as total cholesterol, triglyceride and low density lipoprotein cholesterol(P<0.05);while high density lipoprotein cholesterol decreased at 12 week(P<0.05).③At 8,12 week,in high fat-fed group serum adiponectin decreased than that in control group(P<0.05),and meanwile the difference of AR1mRNA between two groups has no statistical significance but the difference of AR1 protein between two groups achieved statistical significance.④The AR1 protein in liver was correlated positively with HOMA-IR,negatively with the serum adiponectin.⑤We can see hepatic steatosis and mononuclear cell infiltration in hepatic lobule in high fat-fed group.Conclusions①We established a sort of impaired glucose tolerance with insulin resistance animal model in Wistar rats by high fat-fed diet.The success rate of this model arrived to 69.2 %.②Non-alcoholic steatohepatitis occurred during progression of IGT.③Serum adiponectin cut down while the expression of AR1 protein in liver increased during the period of IGT. Adiponectin can be used as an early figure to predict non-alcoholic steatohepatitis.Adiponectin and AR1 can be used as molecule target to do intervention treatment in IGT and predict non-alcoholic steatohepatitis. |
