Objective: Ductus choledochus can induce multiple organs injury. At present, a plenty of animal experimentation and clinic research study on liver, kidney, heart damage mechanism and pathophysiology . But, there are not report about cholecystitis with ductus choledochus block and bile bacteriology serum and gallbladder tissue oxygen free radicals change. The present study development the animal model of acute ductus choledochus block. Through observing block different time and after release block gallbladder pathological, bile bacteriology serum and gallbladder tissue nitric oxide(NO), Superoxidedismutase(SOD), Malonbialdehyde change . After that, we hope that we can understand the disease mechanism deeply and offer the theory support for cure ductus choledochus block induce cholecystitis at clinic.Methods: 72 healthy rabbits weighing 2.0-2.5Kg were randomly divided into 9 groups equally; The normal control group (n=8), the group of bile duct ligation one day(BDL1) (n=8), the group of bile duct ligation three day(BDL3) (n=8), the group of bile duct ligation five day(BDL5) (n=8), the group of bile duct ligation seven day(BDL7) (n=8), treating group: relieve ligation after bile duct ligation one day(T1) (n=8), relieve ligation after bile duct ligation three day(T3) (n=8), relieve ligation after bile duct ligation five day(T5) (n=8), relieve ligation after bile duct ligation seven day(T7) (n=8). All the rabbits were prohibited to eat but not prohibited drink before operation. After build animal model succeed , the group of bile duct ligation rabbit were killed after ligation one day, three day, five day, seven day. The treating group animal were killed after relieve ligation three day. The following studies were performed in every group: taking 2ml bile make bacteriology develop. Spectrophotometric method was used to estimate NO, SOD, MDA in blood and gallbladder tissue. The gallbladder pathologic changes were observed with light microscope after HE staining. The experimental data was expressed as mean+standard deviation. The statistic significance were analyzed by SPSS 13.0 a statistic sofeware.Results: Ductus choledochus induce cholecystitis, earlier period of bile ductus ligation, BDL1, BDL3 group, gallbladder intumesce, gallbladder paries, edema, incrassation, membrane plica integrated, light microscope: membrane epidermis integrated, inherent membrane, muscle, serosa edema, blood vessel dilate, congestion, and a few inflammatory cell soakage(BDL1, BDL3 group). Along with ligation time prolong, BDL5 group gallbladder inflammatory aggravate , gallbladder intumesce, floccule soakage, paries become incrassation, bile thickness , membrane accrete ash yellow exudation or come into being sham flime, part membrane putrescence desquamate, froming small ulceration . light microscope: membrane epidermis desquamate, blood vessel dilate, congestion, membrane, inherent membrane, muscle, serosa have a plenty of white blood cell soakage. BDL7 group, gallbladder nigrescence, paries become weak, membrane plica disappear, take on putrescene, light microscope: membrane epidermis disappear, a plenty of white blood cell oddment and bleed, muscle layer slur, often concomitancy gallbladder around inflammatory, when inflammatory become severity, gallbladder may mortification. BDL5 BDL7 often along with gall-stone.Gallbladder tissue and blood serum NO MDA gradually goes up with ligation time prolong, and lifted up to top level in the group of bile duct ligation seven day(BDL7), Compared with every the group of bile duct ligation was significant difference(P<0.05). Gallbladder tissue and blood serum SOD gradually goes down with ligation time prolong, and reduced significantly in the group of bile duct ligation seven day(BDL7), Compared with every tthe group of bile duct ligation was significant difference(P<0.05). Treating group, the levels of NO,MDA in gallbladder tissue and blood serum were reduced significantly than the group of bile duct ligation , SOD increase compared with BDL group, there was no significantly difference in every group(P > 0.05). After bile duct ligation, the number of bacteriology in bile was increase, after relieve ligation, the number of bacteriology in bile was reduce.Conclusions: 1. Bile duct ligation induce cholecystitis, earlier period of bile ductus ligation, gallbladder intumesce, gallbladder paries, edema, incrassation, membrane plica integrated, Along with ligation time prolong, gallbladder inflammatory aggravate , gallbladder intumesce, floccule soakage, paries become incrassation, bile thickness , membrane accrete ash yellow exudation or come into being sham flime, part membrane putrescence desquamate, froming small ulceration . After relieve ligation, gallbladder inflammatory lighten. 2. After bile duct ligation, Gallbladder tissue and blood serum NO MDA gradually goes up with ligation time prolong, SOD gradually goes down. Treating group, the levels of NO,MDA in gallbladder tissue and blood serum were reduced significantly than the group of bile duct ligation , SOD increase compared with BDL group, The changes of oxygen free radicals play an important role in the happening , developing of bile duct ligation induce cholecystitis. 3. After bile duct ligation, the number of bacteriology in bile was increase, after relieve ligation, the number of bacteriology in bile was ireduce.
