Objects:To study if there is response of P-glycoprotein in microvessel endothelial cells of blood-brain barrier to AEDs . Select VPA and LTG , which are commomly used AEDs , to be added on rat brain microvessel endothelial cells for 8 days. Compare action of P-glycoprotein before and after adding AEDs. Compare action of P-glycoprotein in brain microvessel endothelial cells between adding VPA and LTG.Methods:1. Segregate and cultivate rat BMEC.2. Identify brain BMEC in immunohistochemistry.3. Devide BMEC into three groups: control group, valproate sodium group and lamotrigine group. According to previous experiment results, valproate and lamotrigine were in effective blood drug concentration . After 8 days, determine action of P-glycoprotein on BMEC by flow cytometer.4. Compare action of P-glycoprotein in BMEC between control group and interfere group.Result:1. Keeping interfering in VPA of effective blood drug concentration result in up-regulation of action of P-glycoprotein in BMEC.2. Keeping interfering in LTG of effective blood drug concentration result in up-regulation of action of P-glycoprotein in BMEC ..Concentration:1. To some extent, action of P-glycoprotein reflects the function of BBB. Primary culture ??of BMEC acts as effective tool of studying BBB in vitro.2. BBB isn't a static lipid membrane barrier, just in conventional opinion. Inversely, it's a self-regulation barrier system.3. Flow cytometer, a sensitive apparatus, facilitate determination of action of P-glycoprotein in BMEC.4. Both VPA and LTG can result in up-regulation of action of P-glycoprotein in BMEC.5. Long-term application of AEDs could be the cause of drug-resistance epilepsy.
