Effects Of Valproate And Lamotrigine Acid On Estrous Cycle, Serum Sex Hormone Levels And Ovarian Morphology In Female Rats With Epilepsy

To explore changes of estrous cycle and ovarian morphology and detect the serum concentrations offolliele stimulating hormone(FSH)，Luteotropic hormone (LH)，estradiol(E2), Progesterone(P) andTestosterone(T)of epileptic female rats induced by Kainic acid(KA))when treated with valproate acid(VPA) and lamotrigine(LTG).288-week-old Sprague-Daweley rats were divided randomly into four groups.Control group feednormal saline2ml every day，rest groups were set up epileptic modle induced by Kainic acid(KA)，randomly administered with VPA、LTG and NS by intragastric administration for30days．Weight per3days，dose adjusting according to body weight．Estrous cycles were monitored with daily vaginalsmears．After30days，Blooding，separating of ovary organization in their diestrus．Ovarian morphometricanalyses were made. Use the electrochemical luminescence method to determine serum FSH，LH，E2，P andT level.VPA group and NS group rats lost ovarian periodicity and first displayed significant change(P <0.01)．VPA group displayed significant weight gain increase (P <0.01)． The level of LH was increased inthe VPA and NS groups compared with control group(P<0.05).The level of FSH was decreased in the VPAgroup compared with the control and LTG groups(P<0.05)．the level of T was increased in the VPA groupcompared with LTG and control groups(P<0.05).the level of P was decreased in the VPA group comparedwith control、LTG and NS groups(P<0.05)．The ovarian in VPA group and NS group exhibited polycysticchanges．The results of HE staining showed that compared with control group，VPA group and NS groupfemale rats had enlarged ovaries with numerous growing follicles, especially many preantral follicles, andthe total number of folliles and atretic follicles increased (P<0.05)．Ultrastructure of growing follicles ofVPA group rats exhibited increase of apoptotic granulose cells，theca cell hyperplasia and more secretorygranules can be found in theca cells.Valproic acid sodium easy cause sex hormone secretion disorder compared withlamotrigine．Epilepsy state affect the rat mature follicle formation and ovulation function and result inpolycystic ovary．VPA intervention can aggravate epileptic rats polycystic ovary change and growingfollicles showed more apoptosis and increase of the secretion in the theca cell hyperplasia.
