Object: To seek the dependablity of Uigur Cervical carcinogenesis on RASSF1A gene hypermethylation in order to establish the foundation for furthermore research on the role played by anti-oncogene methylation in tumor generation course and provide theory basement for earlier period clinical diagnosis and demethylation etc. tumor therapy, we made the study.Methods: Methylation-specific PCR (MSP) assay was used to detect the methylation status of promoter of RASSF1A gene in 65 cervica1 carcinoma and 40 norma1 cervica1 tissues.We made statistical analysis to compare the methylation rates in two groups and analyzed the methylation status of the tumor suppressor gene RASSF1A's promoter in Uigur Cervical Squamous Cell Carcinomas.We also made correlation analysis between RASSF1A gene methylation and Cervical Carcinomas lymphoid node metaptosis.Results: Hypermethylation of RASSF1A was observed in 13 of 65 cervica1 cancer cases(20%).Among them 4 cases show homomethylation and 9 cases show heteromethylation. The rest cancer cases and none of norma1 cervica1 tissue was found to be methylated.Statistical analysis indicates that RASSF1A gene methylation status is different in Uigur Cervical Squamous Cell Carcinomas to Uigur Cervical normal epithelium tissue groups(P<0.01).At the same time we did not find statistical significance between RASSF1A gene methylation and lymphoid node metaptosis. We also foud that there is no statisticsal difference of RASSF1A gene methylation rates in low grade clinical stage (6.7%,1/15)and high grade clinical stage (27.0%,10/37)tumor.Conclusion: (1) RASSF1A gene inactivation caused by it's promoter methylation may take part in Uigur Cervical Squamous Cell carcinogenesis. (2)It is possible that RASSF1A gene promoter methylation neither is relative to lymphoid node metaptosis nor to tumor clinical stage.
