Phosphatidylcholine (PC) is a kind of phospholipids, which has special characteristics and physiological function. It was widely utilized in the field of health product,cosmetic,food and pharmaceuticals industry. In particular, it is promising to prepare liposomes and pharmic emulsifier. A simple rapid and accurate quantitative method was established for the determination of phosphatidylcholine by HPLC-UV in this research. Separation was obtained by using lichrospher Si (4.6×250mm, 5μm) column and mobile phase of n-hexane/2-propanol (1:1,V/V) and n-hexane/2-propanol/1% acetic acid (4:4:1) with flow rate of 1.0mL/min,detected wave was 205nm. Under this condition, PC and PE were well separated in 25min. Alcohol soluble phospholipids was detected contenting PC 40.08%(wt%) and PE 19.1%(wt%) .In order to improve the PC content, a study has been started from alcohol soluble phospholipids with using macroporous ion-exchange resin S. Though experiments on S, the optimal conditions were established: adsorption with PC concentration 3mg/mL and 1mL/min, desorption with 95% ethanol and 1mL/min. The product was light yellow, 94.02% PC and the yield was 80.74%.Adsorption dynamics research of PC indicated that adsorption process could be described with shrinking unreacted core model. The adsorption rate of PC was governed by intra-particle diffusion and adsorption time t was in the good linear relationship with 1 ? 3(1 ? X ) 2/3+ 2(1 ? X). The adsorption isotherm of PC could be described with the equation of Langmiur, adsorption reaction was exothermic reaction at the same time.For comparing with resin column, it is also used Al2O3 column to prepare PC, eluting with 95% ethanol and the product had 92.7%PC.The result of comparing the two PC products as follows: the two both had palmitic acid,palmitoleic acid,stearic acid,oleic acid,linoleic acid,linolenic acid, and the content of linoleic acid was at the most; the outcome of HPLC-MS analysis was the product had different molecule species, such as: 16:1/18:1-PC,16:0/18:2-PC,18:2/18:2-PC,18:1/18:2-PC,18:0/18:2-PC and 18:2/18:3-PC, the maximum was 18:2/18:2-PC.
