| Lotus leaves, leaves of the plant which belong to Nelumbo of Nymphaeaceae, produced mainly in Hu Nan, HU Bei, Zhe Jiang and Jiang Su etc province, has various efficacy such as clearing summer-heat, reducing weight and fat, elevating yang and cooling blood to stop bleeding, and is the valuable plant resources which has dual purpose can be used as medicine and food in the No. 51 document which issued by the National Ministry of Health in 2002. The utilization degree of lotus leaves is very low although it has rich resources in china, and it is often used as decoction of Chinese herbal medicine and others excipient of health products by collecting the naturally exfoliatived dry leaves. With the rapid development of the social productivity level , science& technology, industrialization and informationization, human health is facing so many pressures which from living environment and life style, so, the plant extract industry which aim to improve the human-health and living quality has become a rising industry in the whole world. Some researches showed that the flavonoids and alkaloid are the two main functional constituents in lotus leaves. The deep exploitation increased as people know more about the health protection function of lotus leaves, but most of them pay close attention to raw material processing and analyzing the chemical material component and content in lotus leaves, the pharmacological functions of LLE, the method to extract flavonoids and alkaloid under experiment condition etc. The research depth and extent is not enough at all. The traditional separation and purification method always depend on organic solvent ,it always lead to solvent residues, lack of safety, so seeking the green and saving energy separation and purification technology is the key problem to efficiently and synthetically exploit the value of flavonoids and alkaloid in lotus leaves. Macroreticular resins extraction method is a modern separation and purification method developed in recent years, it became popular because it had some advantages such as lower cost, simple equipment, saving energy and reproducible as well, According to the present condition of flavonoids and alkaloid exploitation and utilization in lotus leaves , this paper took the dry lotus leaves as material, studied the several macroreticular resins extraction methods systemically.The results are as following:1,The research compared the effect of 5 kinds of resins of X-5,HP-20, NKA,AB-8 and HPD-100. The results showed that HP-20 was the best one. Its static absorbtion was 68.90 mg/g resin. The rate of desorption was 87.23 %. The research optimized the technique parameters systematically of HP-20 separating lotus flavones. The results showed that the saturated absorption of lotus flavones by HP-20 was 15 BV of lotus flavone of 7 mg/ml concentration. The concentration of lotus flavones could reach 62.06 % desorbed by 40 % ethanol for one times when the concentration of the added lotus flavones was 200 mg/ml and the speed of desorption was 2 BV/h. The rate of lotus flavones desorbed by 40 % ethanol was 81.40 % and the total rate of desorbed lotus flavones was 98.17 %. The concentration of lotus flavones can reach to 98.87 % if they are separated for several times.2,By using the lotus leaves as raw material, after extracting alkaloids from lotus leaves by 30% ethanol, the effect of static adsorption and resoluting of macroporous resin -PA(13-40),D101 and HP-20 for lotus alkloids is compared .Then on the basis of screening the best macroporous resin with higher adsorbing rate and resoluting amount, the effect of the resoluting quantity, purity and yield rate of alkloids at different eluting velocity by the gradient of different eluting density of ethanol whose volume is 3BV is studied ,and the effect of resoluting quantity,purity and yield rate of alkloids by the same gradient of different eluting density under the best eluting velocity is also studied. The results show that D101 is the best resin among the three kinds of resin. When to separate and purify lotus alkaloids by D101,the parameters of 1.5BV/h,3BV resoluting volume of each ethanol concentration and 20%,40%,60%,80% ethanol gradient resoluting are the best ,and the total quantity of eluted alkaloid, yield rate, the total purity can reach 10.8226mg,62.9%,4.86% in quence , furthermore ,when only collecting the resoluting liquid of 80% ethanol, the purity of lotus alkaloids can reach 32.56%,which is valuable for preparing the higher purity lotus alkaloids.As stated before, this study beginning from analyzing the problems existed in extraction methods of flavonoids and alkaloid in lotus leaves, compared several big-poled separation technologies systemically ,got technical parameters of macroreticular resins extraction method ,put forward the data and science theories for lotus leaves utilization efficiently and synthetically. |
