ObjectiveTo investigate the role of endoplasmic reticulum apoptosis in the process when proteasome inhibitor MG-132 inhibiting proliferation and inducing apoptosis of human gastric cancer cell line SGC7901.MethodsMG-132 was prescribed for human gastric cancer cell line SGC7901,the cell-growth inhibitory effect was observed by MTT assay,the mark of the endoplasmic reticulum apoptosis in gastric cancer cells induced by MG-132 was detected by FCM(flow cytometry), apoptotic bodies were found in cells by scanning electron-microscopic examination..ResultsMG-132 can significantly inhibit the proliferation of SGC7901 cell.IC50 of 24h,48h and 72h was 80.767μmol·L-1,8.596μmol·L-1,0.509μmol·L-1.The confidence interval of 95% was 54~383μmol·L-1,6.107~13.838μmol·L-1 and 0.250~0.801mol·L-1. There was obviously expression of mark of the endoplasmic reticulum apoptosis in gastric cancer cells induced by MG-132 after 48h and a great quantity apoptotic bodies were found in cells by scanning electron-microscopic examination..ConclusionsThis study proves that proteasome inhibitor MG-132 can inhibit proliferation and induce apoptosis of SGC7901 cell significantly, and force cell accessing to the endoplasmic reticulum apoptosis pathway.
