Objective T o investigate the clinical and pathological relationship of TGFβ1-509C/T polymorphism with ANCA associated systemic vasculitis with nephrosis in han Nationality of Guangxi Population.To ensure the susceptible gene of ANCA associated systemic vasculitis,and illustrate the causes of ANCA associated systemic vasculitis from gene level.Methods The genotypes of TGFβ1 gene polymorphism(-509C/T)was determined by polymerase chain reaction-restriction fragment-length polymorphism analysis in 80 normal control subjects and 36 ANCA associated systemic vasculitis in the case-control study in Chinese people in guangxi region.At the same time,to obtain general clinic information,such as blood pressure,blood fat,renal function,ANCA, renal pathology and so on.Result 1.TGFβ1 gene(-509C/T)include 3 genotype(CC,CT,TT)and 2 allele gene(C or T)in han Nationality of Guangxi Population.2.There were no statistical significant difference of genotypes frequency[CC,CT,TT=20%,60%,20%(NC group)VS.22.2%,61.1%,16.7%(AASV group),χ~2=0.2098,P>0.05]and allele frequency[C,T=50%,50%(NC group)VS.52.8%,47.2%(AASV group),χ~2=0.1533,P>0.05]of -509C/T polymorphism in TGFβ1 gene between ANCA associated systemic vasculitis and normal control subjects.3.There are statistical significance between TT genotype and the other two genotypes about IgM level(P(TTvsCT)=0.021,P(TTvsCC)=0.019),systolic blood pressure(P(TTvsCT)=0.010,P(TTvsCC)=0.040),the frequency of ANCA is different statistically between TT genotype and the other two genotypes(P(TTvsCT)=0.0384,P(TTvsCC)=0.015),and there are statistical significance between CC genotype and the other two genotypes about 24hUPro(P(CCvsCT)=0.021,P(TTvsCC)=0.016).No correlation was found in the -509C/T Polymorphism of TGFβ1 with sex,age,gross hematuria,BUN,Scr,ESR and ENA(P>0.05),etc.4.The CC genotype frequency was higher in pations with Med+Heavy degree mesangial proliferation corresponding TT genotype decreased (P<0.05).No correlation was found in the -509C/T Polymorphism of TGFβ1 with other groups of renal pathology,Renal pathology immunofluorescence sedimentum.Tubulointerstitial Lesion score was statistically higher in pations with CC genotype than those with TT genotype and CT genotype,P(CCvsCT)=0.022,P(CCvsTT)=0.022。conclusion 1.TGFβ1 gene include 3 genotype(CC,CT,TT)and 2 allele gene(C or T)in Chinese ban people in GuangXi Population.2.-509C/T polymorphism in TGFβ1 gene was not possiblely associated with susceptibility to ANCA associated systemic vasculitis with nephrosis in Chinese han people in GuangXi Population.3.-509CC genotype in TGFβ1 may be associated with susceptibility to heavy albuminuria,Med+Heavy degree mesangial proliferation and Tubulointerstitial Lesion score of ANCA associated systemic vasculitis with nephrosis in Chinese han people in GuangXi Population.4.-509TT genotype in TGFβ1 may be associated with hemocircular low IgM level,high systolic blood pressure and the production of PR3-ANCA.
