Studies On Immunopotentiating Activity And Effects Of Antibody Response To Chinese Herbal Medicinal Polysaccharides

In order to screen the novel immunopotentiator from Chinese Herbal Medicinal polysaccharides(CHMPs),at the same time investigated their application in clinic,Isatis root polysaccharide(IRPS),Chinese yam polysaccharide(CYPS),Astragalus polysaccharide(APS)and Achyranthes bidentata polysaccharide(ABPS)were extracted by Water-extracting and ethanol-precipitation method from four kinds of Chinese Herbal Medicines(CHMs)which were Isatis root(IR),Chinese yam(CY),Astragalus(A)and Achyranthes bidentata(AB).Effects of the four polysaccharides on the specific and non-specific immuno-factors and immunocytes in vivo and were studied and their immunopotentiating activity in clinic were compared.Then these CHMPs could be selected as the novel immunopotentiator.The five experiments were done.Experiment 1:The four polysaccharides including IRPS,CYPS,APS and ABPS were extracted by Water-extracting and ethanol-precipitation method from IR,CY,A and AB.The contents of four polysaccharides were measured by Vitriol-anthracene ketone,and the net contents of every polysaccharides are 58%,73%,65%,54%.They were used as the materials of the following experiments,and what's more,The study made the foundation for the whole study.Experiment 2:In order to screen the novel immunopotentiator from CHMPs in mice,The assays of peritoneal macrophage,hemolysin and hemolysis void in mice were carried out.The effects on immune system in mice to the four CHMPs were studied.The results showed that IRPS,CYPS,APS,ABPS all could increase cellular immune function of mice,enhance the phagocytic activity of peritoneal macrophage and increase the formation of hemolysin and hemolysis void in mice compared with the control group.It implied that IRPS,CYPS,APS,ABPS all could increase the humoral immune level of mice.Experiment 3:In order to select CHMPs that had immunocompetence and developed a new-type immunopotentiator used by swine,Effects of the four CHMPs on proliferation of swine splenic lymphocytes were investigated by the methods of MTT colorimetric assay.The results showed that IRPS,ABPS,CYPS and APS could promote proliferation of swine splenic lymphocytes significantly(p<0.05),which were induced by ConA or LPS,and the effects were related to concentration of the polysaccharides.Experiment 4:In order to determin the optional concentration of the four CHMPs, the experiment of the swine splenic lymphocytic transformation was carried out.The results showed that IRPS,APS,CYPS and ABPS could promote proliferation of swine splenic lymphocytes which were direct or induced by ConA,LPS significantly(p<0.05);the function that promoted swine splenic lymphocytic proliferation was related to concentration of polysaccharides,and the optimal concentration was different.Experiment 5:In order to study immunopotentiating activity of CHMPs in clinic, Healthy young swine were injected respectively with the four CHMPs,at high and low dose,once a day for three successive days after they were immunized with the vaccine of PRRSV,then we determine the antibody level by ELISA test kit.It was found that the four CHMPs would be arranged in IRPS,CYPS,APS,ABPS according to their immunopotentiating activity.The results showed that the four CHMPs could increase porcine special antibody response significantly.These experiments indicated that IRPS,CYPS,APS,ABPS were able to activate immune function of mice,promote proliferation of swine splenic lymphocytes,and increase porcine antibody level.These studies showed that the four CHMPs could promote the specific and non-specific immuno-factors and immunocytes in vivo and, and the studies would offer scientific basic for clinic application of these CHMPs.
