Experimental Study On The Effect And Mechanism Of SOD-Liposome Treating The Rats Skin Injury Induced By β Radiation

Objective: To investigate the effect of SOD-Liposome treating radiation skin injury and the mechanism of SOD-Liposome promoting the ulcer healing on the base of establishing The animal model ofβradiationⅡdegree injury model.Methods: 54 SD rats are randomly divided into 3 groups:A.15Gyβradiation,n=14;B.45Gyβradiation with application of SOD-Liposome after the skin ulcer appear, n=20;C.45Gyβradiation with application of normal saline after the skin ulcer appear,,n=20.The healing time and the number of healing rats were observed, parts of the ulcer tissue were taken respectively on the 0th,1th,2th,3th week after the ulcer had formed.The pathological changes were observed macroscopically by ligh and electron macroscopy . MDA,the contents of total collagen,collagen type I and typeⅢin the tissue were measured.Results: 1. The model ofβray radiationⅡdegree skin injury on rats was established successfully under 45Gy. 2. Compared with the control group , the ulcers'healing time was shorter and the number of healed rats was obviously larger. 3. The fibroblast amplification is active, altering the maturation, secrete prosperous, but the aging delay.4.The content of the free radicals in the tissue of ulcers reduced after application of the medicine,but didn't reduce contineously. 5. Compared with the control group, the treating group contented more collagen of typeⅢand little of typeⅠ,the total amount of collagen of two group have no difference.Conclusion: 1. The animal model ofβradiation skin injury ,produced by the linear accelerator, is easy to establish and the dosage is accurate.2. The exernal application of SOD-Liposome can promote healing of radiation skin injury in rats nd shorten the healing time. 3. The mechanism is maybe reducing the free radical level of skin ,easing the hurt to fibriblast,promoting damaged fibroblast functional rehabilitation and strength collagen protein synthtase,particularly the secret of typeⅢcollagen,thus be advantageous to the healing of ulcer.
