Objective:①To extracte and separate hydroxysafflor yellow A (HSYA)from Carthamus tinctorius L.,and use it as reference substance in qualitative and quantitative identification.②To analyze the quality of Carthamus tinctorius L.in Emin Tacheng Xinjiang,and study the factors that effect its quality.③To establish chromatographic fingerprint of Carthamus tinctorius L.in Emin and analyze it.④To investigate and summary habitat environment of Carthamus tinctorius L.in Emin.Methods:①Extracted and separated HSYA from Carharmus tinctorius L.by using D-4020 macroporous resin column chromatography and sephadex LH-20 chromatography.②Detected the content of HSYA in Emin Carthamus tinctoius L.by using the HPLC method of Chinese pharmacopoeia(2005).The C18-ODS(250×4.6mm)column and methanol-acetonitrile-0.7%H3PO4(26:2:72)as a mobile phase were used. The flow rate was 1.0ml/min,the column temperature was 25℃,and the detective wavelength was 403nm.In this way,the effects of such condition as varieties,collecting time and storage time were studied.③The gradient elution mode was applied in establishing HPLC fingerprint of Emin Carthamus tinctrius L,and data was analyzed by "system of traditional Chinese medicine analyzing and data managing" software.Compared and analyzed the difference fingerprint between two varieties of high quality Carthamus tinctrorius L.in Emin.④Collected and summarized the information of growing environment in Emin.Results:①The purity of HSYA that extracted from Carthamus tinctorius L.was more than 98%, and it could be used as reference substance in content determination.②The experiment indicated,the content of HSYA in Xinhong-5 was the highest in these detected varieties,the optimal time to pick safflower was the morning of the third or forth day after flowering,storage time could affect the quality of safflowers.③Established the fingerprint of high quality safflower in Emin.There were evident difference between them in despite of same growing environment.Conclusion:①The purity of HSYA was 98%,it was higher than the earlier result of 93%.②All these work of quality studying and environment investigating were the basic study for GAP(Good Agriculture practice)of Emin's safflower,and may provide some guide for growing,collecting and storage of local safflower.③The fingerprint of Emin safflowers reflect the intrinsic quality completely and provide comprehensive methods of identification and quality evaluating for local safflower.It is necessary for the modernization of traditional Chinese medicine.
