| Objective Cell division and embryonic development are subject to be inhibited in spontaneous abortions. We approach the expression of spindle checkpoint genes(MAD2 and BUB1) related closely to cell cycle regulation in spontaneous abortions, so as to further find out the development mechanisms for causing spontaneous abortions. It will establish important foundation in searching new diadynamic criteria for laboratory diagnosis in spontaneous abortions.Methods:1 MAD2 and BUB1 mRNA levels were compared by using FQ-PCR in chorionic tissues of embryo from induced abortions and spontaneous abortions.2 MAD2 and BUB1 protein levels were determined by using western blot。Result:1 Mean MAD2 mRNA/GAPDH mRNA was 0.00941±0.00026 in chorionic tissues of embryos from induced abortions(control group) and 0.00835±0.00041 in spontaneous abortions(experimental group), meanwhile, it was 0.00879±0.00035 in once spontaneous abortions and 0.00901±0.00033 in twice or more spontaneous abortions . Mean BUB1 mRNA/GAPDH mRNA varied from 0.00671±0.00021 in control group to 0.00597±0.00036 in experimental group,it was 0.00682±0.00025 and 0.00713±0.00041 in once spontaneous abortions and twice or more spontaneous abortions correspondingly.2 Mean MAD2/β-actin optical density was 0.5790±0. 0331 in chorionic tissues of embryo from induced abortions and 0.2683±0.0275 in spontaneous abortions,in addition, it was 0.2791±0.0311 and 0.0431±0.0020 separately in once spontaneous abortions and twice or more spontaneous abortions. Mean BUB1/β-actin optical density was 0.6260±0.0209 in induced abortions and 0.3033±0.0406 in spontaneous abortions, therefore it was 0.3153±0.0027 and 0.07122±0.0030 in once spontaneous abortions and in twice or more spontaneous abortions.3 By statistical analysis, mRNA level of MAD2 and BUB1(p>0.05) in induced abortion group and spontaneous abortion group have no statistics difference ,but their protein levels between two groups have statistics difference(p<0.01) , protein level was significantly higher in once spontaneous abortion group than twice or more spontaneous abortion group as well.Conclusion: The protein expression of MAD2 and BUB1 are distinguished lower in experimental group than that of in control group ,but their mRNA levels have no difference .Reduction of MAD2 and BUB1 expression in spontaneous abortions indicate that they may play important roles in cell division of embryo and embryonic development. |
