| AIM:To build the model of Yin deficiency rats with hydrocortisone, and observe the effects of Nourishing Yin Antitoxic Capsule 2 on the corticosteroid level of Yin deficiency rats, and the expression of Glucocorticoid receptor proteins and mRNA in the liver of Yin deficiency rats, and investigate how the Nourishing Yin Antitoxic Capsule 2 affects the function of hypothalamic-pituitary-adrenocortical (HPA)axle.Methods:Forty SD rats were randomly allocated to five groups: normal control group, Yin deficiency model control group (YD), Nourishing Yin Antitoxic Capsule 2 group (YC2), Liuwei Dihuang Pills group (LD) and Guifu Dihuang Pills group( GD ) . Radio-immunity method was used to observe the contents of corticosteroid (CORT) of these rats. Immunohistochemistry was used to observe the expression of Glucocorticoid receptor proteins in the liver cell nucleus. RT-PCR was used to observe the level of Glucocorticoid receptor mRNA in the liver cells.Results:Compared with the normal group, the contents of CORT were higher in YD group (P<0.05), which indicated that the model of Yin deficiency rats were successfully built; Compared with YD group, the contents of CORT in YC2 group and LD group were lower (P<0.05), which indicated that both of them could inhibit the CORT-increasing function of GC; And compared with YD group, the contents of GD group was higher (P<0.05), which indicated GD could not inhibit the CORT-increasing function of GC.Compared with the normal group, the expression of GR in the liver cell nucleus in YD group was lower (P<0.05); The expression of GR in the liver cell nucleus in YC2 group was higher than that of YD group (P<0.05), but still lower than that of the normal group (P<0.05); The expression of GR in the liver cell nucleus in LD group was higher than that of YD group (P<0.05), but had no difference with YC2 group (P>0.05); The expression of GR in the liver cell nucleus in GD group was lower than that of the normal group (P<0.05), but lower than than that of YD group, which had no sense in statistics (P>0.05).Compared with the normal group, the level of GR mRNA in YD group was lower (P<0.01); Compared with YD group , the level of GR mRNA in YC2 group was higher (P<0.01); Compared with YD group , the level of GR mRNA in LD group was higher (P<0.01), but was lower than that of YC2 group (P<0.01); Compared with YD group , the level of GR mRNA in GD group was higher (P<0.01), but was lower than that of YC2 group and LD group. Conclusions: In this experiment, the contents of CORT in YD group were higher than in the normal group, and the expression of GR protein in the liver cell nucleus was lower, and the level of GR mRNA in liver cells was also lower. Nourishing Yin Antitoxic Capsule 2 could decrease the contents of CORT, improve the expression of GR and the level of Glucocorticoid receptor mRNA, which all might be one of reasons why Nourishing Yin Antitoxic Capsule 2 could prevent and cure the side-effects of Glucocorticoid. |
