α-linolenic acid, scientific name is 9,12,15-oc-tadecatrienoic acid, cis form, non conjugate solid configuration, its molecular fomula is C18H30O2, belongs to n-3 unsaturation fatty acid series, it's the necessary fatty acid of human being. The latest researches shows that Perilla frutescens is the plant contents abundant ofα-linolenic acid, perilla frutescens seed oil is light yellow oleaginous liquid, contentsω-3 unsaturation fatty acid series, exists as eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) after being metabolized. EPA is the precurosor of rostaglandin-triene(eg PGI3,TXA3 ) , DHA is the main components of phospholipid in nervous system of brain and retina, both of them have great physiological functions. Besides the physiological functions above,α-linolenic acid also has many pharmacologic actions, such as decreasing blood fat and blood pressure, restraining hypersensitiveness, anti- thrombus, it also has some function of anti-cancer, it can restrains some tumor obviously. So we can exploiture it into therapeutical drugs or assistant therapeutical drugs that apply to many chronic-diseases,such as brain epistom,high cholesterol-emia,hign blood pressure,myocardial infarction,asthma and cancer.Because of containing manyα-linolenic acid, the oxidation stability and heat stability of perilla frutescens seed oil is not good, so it influences the application of perrila frutescens seed oil , so some experts are studying of making it into powder and ointment, in order to make it stable . China is at the underway phase of studying of the dosage forms of perilla frutescens seed oil, there are little products on sale, only soft elastic capsule of it has been reported. So , on the foundation of reading many literatures, we choose the intravenous emulsion to be the dosage form of perilla frutescens seed oil. At first common lipid emulsion is blank, it can offer nutrition for patients who have been operated just now. With pharmacy developing, the lipid emulsion became a new carrier of indiscerptible dugs. It not only can improve stability, but can avoid bring more organic impregnant. At the same time concentration of medicine in out-water-phase is very low, it can reduce possibility of phlebitis. In addition intravenous emulsion has strong target ability. It can arrive at the RES of liver, kidney, spleen, lung, heart and marrow. So concentrationg of other tissues are lower correspondingly. Evaluate the intravenous emulsion through its appearance,particle size and distribution and centrifugal stability constant, the basel prescription of intravenous emulsion and the contents of every component were decided using single factor method, then using orthogonal experiment method we decide the best prescription : perilla frutescens seed oil 1.5%, Refined egg phospholipid 1.6%, refined intravenous bean oil 6%, oleic acid 0.04%, VE 0.03%, glycerol 2.5%, the rest is intravenous water. By testing of particle size and the appearance, decided the first temperatue for original emulsion is 80℃,the rotate speed is 19000rpm and the temperature before flow into the high pressure homogenizer is 40℃using the orthogonal experiment method, then we decided secondary P 1000bar and first P 100bar ,3times. Finally, adjust pH to 8.0 and sterilizing for 15min under 126℃.Establish quality evaluation and content determination method ofα-linolenic acid by gas chromatography(GC). We also inspect the rheological quality, electricity quality and osmotic pressure quality of intravenous emulsion, using influence factor and long time experiments we inspect the stability of intravenous emulsion, the result shows that its stability is accorded with pharmacopoeia.Through injecting three different dosages of intravenous emulsion into big rats, inspected the drug action of it. We founded that this intravenous emulsion can reduce the concentration of cholesterol and triglyceride in the serum of rats., it can also influence the concentration of high density lipoprotein–cholesterol in the serum of rats. The result showed that this product can reduce the concentration of blood fat of rats, the deep researches of its pharmacology and drug action need to do.
