Diabetes Mellitus(DM)is a common chronic disease.In patients with type 2 diabetes has accounted for more than 90%,is the main pathogenesis of insulin resistance and islet(3 cell damage,both influence each other,promote each other,resulting in blood sugar,blood lipid metabolism disorders.Regarding this topic research about the two kinds of pomegranate seed oil,Perilla seed oil,are both highly unsaturated natural oils.Punicic acid is an important physiological active ingredients of pomegranate seed oil,alpha linolenic acid is an important physiological active ingredients of Perilla oil,the content of these two kinds of components in each oil is very high(60%-80%).Punicic acid is highly unsaturated and omega-3 type fatty acids,which physiological function is the same as alpha linolenic acid,as the ideal composition adding the human body lacking of omega-3 polyunsaturated fatty acids.In recent years,the study found that the vegetable oils rich in polyunsaturated fatty acids are of very beneficial physiological functions,including anti-cancer,falling blood pressure,resisting cancer,weight loss,etc.,especially researches of diabetes and hyperlipidemia has caused more and more attention.For its efficacy in domestic research,in order to have a better use of Perilla and pomegranate seeds and develop a functional food about falling blood sugar and blood lipids,this study aims to compare with pomegranate seed oil,Perilla seed oil and combination of this two kind of oils in falling blood sugar and blood lipids through the establishment of type 2 diabetes animal model simulating human type 2 diabetes mice,and discussing the possible mechanism.In this paper reviewed the present reaearch about the fatty acid composition and pharmacological effects of pomegranate seed oil and Perilla seed oil.On the basis,the study mainly used supercritical CO2 fluid extraction,having a investigation and optimization of the process parameters about pomegranate seeds,Perilla seed and mixture of this two kinds of seeds;Putting Pomegranate acid and alpha linolenic acid as reference substance,the paper tried to establish pre-column derivatization HPLC method for the content determination of extraction of oil;To establish a mice model of type 2 diabetic and observe the effect of pomegranate seed oil and perilla seed oil and blend oils on glycometabolism and lipid metabolism.1.The technical researchUsing supercritical CO2 fluid extraction method extract oils from pomegranate seed,Perilla seed and the mixture seeds of the mass ratio of 1:1.By single factor test,to get the oil rate X(%)and grease pure brightness as examining index,the study determined the best supercritical C02 extraction technology conditions of pomegranate seed oil,Perilla seed oil and blend oils.Getting pomegranate seed oil extracted from pomegranate seed and its optimal supercritical extraction conditions for:extraction pressure for 29-30 Mpa,extraction temperature of 45℃,the extraction time was 1 h;Separation pressure of the separation kettle I is 12Mpa,separation temperature of 50℃;Separate pressure of the separation kettle II is 5 Mpa,separation temperature of the separation kettle II is 35℃.Getting perilla seed oil extracted from perilla seed and its optimal supercritical extraction conditions for:extraction pressure for 29-30 Mpa,extraction temperature of 40℃,the extraction time was 3 h;Separation pressure of the separation kettle I is 12Mpa,separation temperature of 50℃;Separate pressure of the separation kettle II is 5 Mpa,separation temperature of 35℃.After the meal of pomegranate seed,the mixture seeds of the mass ratio of 1:1,grinding into fine powder,Getting blend oils extracted from mixtures of seeds and its optimal supercritical extraction conditions for:extraction pressure for 29-30 Mpa,extraction temperature of 40℃,the extraction time was 2 h;Separation pressure of the separation kettle I is lOMpa,separation temperature of 50℃;Separate pressure of the separation kettle II is 5 Mpa,separation temperature of the separation kettle II is 35℃.2.Quality standard researchWith punicic acid and alpha linolenic acid as reference substance,the study tried to established pre-column derivatization HPLC method for the content determination of pomegranate seed oil,perilla seed oil,the adjustable mixed oil(pomegranate seed oil:Perilla seed oil=1:1)and mixed oil(pomegranate seeds:Perilla seed=1:1).3.Research on the pharmacologyThis part of the study is mainly to observe the effect of pomegranate seed oil and perilla seed oil and blend oils on glycometabolism and lipid metabolism and discusses its possible mechanism through the establishment of the two different experimental type 2 diabetic mice model.At the same time,we evaluated whether the mixed oil of hypoglycemic and lipid-lowering effect is better than the single herbs,thus providing certain reference for pomegranate seed oil and Perilla seed oil formula.3.1 Effect of the compound pomegranate seed oil and perilla seed oil extract in mice with type 2 diabetic caused by high fat diet and streptozotocinThis experiment adopts the high fat feed first,and then in combination with small dose of STZ induced hyperglycemia and hyperlipidemia model.Experiment divided into 7 groups,the normal group,model group,metformin group,pomegranate seed oil group,Perilla seed oil group,the high dose self-regulation oil group,the low dose self-regulation oil group,administrated for 9 weeks.Compared the effect of pomegranate seed oil,Perilla seed oil and the self-regulation oil on fasting blood sugar,TC,TG,LDL-C,uric acid,total bilirubin of type 2 diabetes mice,and discusses its effects on liver apolipoprotein B100,FAS enzyme levels.Results showed that the pomegranate seed oil have fall blood sugar function,Perilla seed oil drop TG effect is better than that of pomegranate seed oil.Self-regulation oil showed better antilipemic activity,but did not affect blood sugar.Compared with the normal group,model group mice has a significant rise in liver Apo B100 and FAS enzyme levels,and statistically difference,which suggesting that diabetes is associated with the rise of this two enzymes.Compared with model group,the liver Apo B100 content in the pomegranate seed oil group decreased significantly,which showed that pomegranate seed oil can inhibit the secretion of Apo-B100,but did not significantly reduced FAS enzyme levels.Compared with model group,the liver FAS content in the Perilla seed oil group decreased significantly,but did not significantly reduced Apo-B100 enzyme levels.Compared with model group,the liver Apo B100 and FAS enzyme levels of high and low dose self-regulation oil were significantly reduced,and high dose effect more apparent.Above results show that pomegranate seed oil,Perilla seed oil may have the different prevention mechanism in type 2 diabetes.3.2 Effect of the compound pomegranate seed oil and perilla seed oil extract in mice with type 2 diabetic caused by streptozotocin and high fat dietThis experiment use STZ intraperitoneal injection and the continuous high fat feed induced experimental type 2 diabetic mouse model.Experiment divided into 9 groups,the normal group,model group,metformin group,pomegranate seed oil group,Perilla seed oil group,the self-regulation oil group,the high dose blend oil group,the low dose blend oil group,administrated for 25 weeks.Compared the effect of pomegranate seed oil,Perilla seed oil and the self-regulation oil on fasting blood sugar,TC,TG,LDL-C,uric acid,total bilirubin of type 2 diabetes mice,and discusses its effects on insulin,liver apolipoprotein B100,FAS enzyme levels at the end of 8 weeks,16 weeks,24 weeks.Results showed the same as the previous.At the same time,the oral glucose tolerance test(OGTT),according to the results of blood sugar,the fasting glucose of normal group mice in 0 min and oral glucose 20,60,120 min were low compared with high-fat model group,which were significant differences in various time points.Compared with model group,the self-regulation oil in 0,60,120 min statistically difference;the blend oil in 60,120 min statistically difference.The mixture of pomegranate seed oil and Perilla seed oil can improve the glucose tolerance of mice with type 2 diabetes.
