Objective To investigate the cytokine( TNF-α,TGF-β1,CTGF)in response for the foeniculum vulgare mill on ccl4-induced liver fibrosis in rats. Methods Liver fibrosis was induced by injecting CCl4 in abdominal cavity and drinking 10% alcohol.70 Healthy Wistar rats were divided into 5 groups randomly, Normal control group (A),Model group (B),Colchicines therapeia group (C, 0.02 mg / day), Foeniculum vulgare mill therapeia group (low, middle, high). Drugs were lavaged for 4 weeks , TNF-α, TGF-β1, CTGF of liver tissue and serum were detected by the method of Immmunohistochemistry and ELISA respectively. TNF-αwas measured by the method of the Enzyme- linked immunospot assay (ELISPOT). Result Morphology stain showed that fatty degeneration and inflammation became light in foeniculum vulgare mill therapeia group(low, middle, high)compared with Model group (B). The immmunohiso- chemistry staining indicated the mount of the cytokines( TNF-α, TGF-β1, CTGF) secreted by the cells could be reduced in C, D, E, F, the number of the positive cells decreased . ELISA's showed the mount of the cytokines( TNF-α, TGF-β1, CTGF) secreted by the cells could be reduced in C, D, E, F. There were significant different between A and B (P<0.05)and significant different among B and C, D, E, F (P<0.05). The high group was better than low and middle group in reducing TNF-α, TGF-β1, CTGF. Using ELISPOT to test the amount of the TNF-αfrom the production of peripheral blood mononuclear cells (PBMCs), There were significant different between A and B (P<0.05) and significant different among B and C, D, E, F(P< 0.05). The high group was better than low and middle group in reducing TNF-α.Conclusion Foeniculum vulgare mill(low, middle, high)can reduce the TNF-α, TGF-β1, CTGF secreted by cells and inhibit the inflammation in rats liver , protect the hepatic cells from damage induced by CCL4 significantly. |
