| Ligustrum lucidum Ait.(LIA) is the dried fruit of Oleaceae plants which is sweet,bitter,cool-nature and belong to the liver and kidney jing point.It can nutritions liver and kidney,improving eyesight and blacked hair.It often be used to treat giddiness,tinnitus,the weak of loin and knee,early whiteness of beard and hair and dim of eyes.The modern research indicate that LIA have the pharmacologic action such as antibiosis,anti-virus,antiinflammatory,lower blood sugar and blood fat,anti-aging,anticancer and increasing immune function and so on.Oleanolic acid(OA) is the main ingredient which can estimate the medical material's quality.As a frequently used medicine in clinic, LIA often used to cure coronaryartery disease,hyperlipemia,hyperglycosemia and hepatitis. LIA have abundant resources in our country,including Eastern China,Southern China, southwest and Central China.LIA like living in the warm and dank places.Shaanxi province(except north area) is one of the natural habits.Our task is develop and utilize orthodox medical material sufficiently.Objective Collecting LIA in different habits and growing stages in Shaanxi province used as researching object.Identification medicinal herbs and determinate oleanolic acid (OA) and ursolic acid (UA) in LIA,in order to evaluating LIA's qualilty,utilizing reasonable and saving resources.Methods1.We picked LIA from five cities(Baoji,Xi'an,Weinan,Hanzhong,Ankang) in Oct. in Shaanxi province,and in Aug.,Sep.,Oct.,Nov. and Dec.,respectively in Xi'an.Removed impurities and storaged the fruits under room temperature as recearching object.2.The paper made comparative research of LIA and its varietics,and depicted the result of morphological and histological characters on crude drugs and some physical and chemical properties.3.By HPLC with Waters600 as its chromatographic system,and Lichrospher C18(4.6mm×250mm,5μm) column was applied with CH3CN-CH3OH-H2O- H3PO4-(C2H5)3N(50∶30∶20∶0.02∶0.04) as its mobile phase,the flow rate was 1mL?min-1.Evaluating LIA's quality in Shaanxi province.Results1.Character identification show that LIA is green and small in its early growing stage.The colour of the fruits changed into henna in Oct.It is ripe when the fruit is purple and black Until Dec.2.In power identification,we can see many epidermic cells,vas,albuminous cells,sometimes can see crystal. It is thus clear in tissue signature that we can see bundles in transections,two-room ovary,one seed in each room,seeds are growing in sequence. 3.Physics and chemistry analysis manifests that LIA contains steroid-Terpene,fats,triterpenoid saponin,and OA from TLC.4.Estabished the method to determinate OA and UA in LIA.Regression equation,OA:Y=12373.29X+108249.93,r=0.9996.UA:Y=12098.02X+85955.73,r=0.9998.OA's linear correlation is favourble in 1.01～5.07μg and UA is 0.41～2.05μg.The recovery rate is 98.8% and 100.5%.So this method is stable and reliable.5.Assaying show:During prolonging growing stages in Xi'an city the highest contents of OA and UA are Oct.(2.61%)and Aug.(1.00%).They both reduced to the lowest point(1.32%,0.55%)in Dec..6.Among different habitats at same growing perieds.The highest contents of OA and UA(3.18%,1.22%)are in Ankang city.Conclusion1.We know the collected LIA are exact.They display common regularity in different growing stages,small difference in micro-identification among diffenent habits.2. During prolonging growing stages in Xi'an city,the highest contents of OA and UA are in Oct.(2.61%)and.(1.00%) in Xi'an city,They both reduce to the lowest point(1.32%,0.55%)in Dec.3.The contents of OA and UA are highest(3.18%,1.22%)in Ankang city among different habitats at same growing perieds.4.It suggested that we should mainly base on the highest contents to select the harvest time according to our demands.In order to save resource,bring into full play maximatily.It provide theoretical foundation,accelarating modernize contribution of Chinese crude drug in Shaanxi province. |
