Background:Ligustri Lucidi Fructus is a kind of traditional Chinese medicine,which can nourish the liver and kidney,improve eyesight and black hair.In clinical practice,processed products are mostly used as medicine.After processing,the property and efficacy of the processing of Ligustri Lucidi Fructus with wine change,and the effect of nourishing liver and kidney is enhanced.However,the chemical constituents before and after processing have not been deeply studied.In this study,the characteristics and biological effects of the processing of Ligustri Lucidi Fructus with wine were studied,and the biological significance of the processing of Ligustri Lucidi Fructus with wine was expounded.Objective:The purpose of this study is to research the chemical constituents and biological effects of the processing of Ligustri Lucidi Fructus with wine.The biological significance of enhancing efficiency and reducing toxicity in the processing of Ligustri Lucidi Fructus with wine was expounded.Methods:The fingerprint of Ligustri Lucidi Fructus and liquor products were established;The characteristic components of Ligustri Lucidi Fructus and processing of Ligustri Lucidi Fructus with wine products were analyzed by UPLC-Q-TOF-MS,and to carry out qualitative and semi quantitative analysis of structure;to explore the mechanism of liver protection,lipidlowering and “toxicity”of characteristic components through network pharmacology;to elaborate the biological significance of the processing of Ligustri Lucidi Fructus with wine.Results:1.Study on fingerprint of Ligustri Lucidi FructusThrough the establishment of the UPLC fingerprint of Ligustri Lucidi Fructus and the processing of Ligustri Lucidi Fructus with wine products,a toal of 35 common peaks were obtained and 2 new peaks were detected.The similarity of 15 batches of Ligustri Lucidi Fructus was between 0.951 and 0.994,which had good similarity with the control fingerprint,indicating that the quality of 15 batches of Ligustri Lucidi Fructus was relatively stable.The similarity of 20 batches of the processing of Ligustri Lucidi Fructus with wine was between 0.766 and 0.998,which indicated that the processing of Ligustri Lucidi Fructus with wine may have some differences due to different manufacturers,operators or operation process,resulting in unstable quality.2.The result of qualitative study on the chemical compents of Ligustri Lucidi Fructus and the processing of Ligustri Lucidi Fructus with wine78 chemical components of Ligustri Lucidi Fructus were identified by High Performance Liquid Chromatography Tandem Quadrupole Timeof-Flight Mass Spectrometry(LC-MS);26 characteristic components of the processing of Ligustri Lucidi Fructus with wine were screened by non targeted metabonomics method,and the structural formulas of 7components were successfully identified;differential analysis showed that18 components increased,8 components decreased after wine making,and2 were new components after wine making.3.Biological effect analysis(1)Biological effects of hepatoprotective effect of characteristic componentsThe results of network pharmacology and molecular docking showed that AKT1,MDM2,Rho A,EGFR,MMP2,PIK3 CA and other targets were the key targets of characteristic components with potential therapeutic effect on liver protection,which might play their potential role by regulating PI3K-Akt signaling pathway,Erb B signaling pathway and other metabolic pathways to play their potential role in liver protection.(2)Biological effect of lipid lowering effect of characteristic componentsThe results of network pharmacology and molecular docking showed that MAPK1、EGFR、MAPK8、MMP9、MTOR and other targets were the key targets of characteristic components with potential therapeutic effect on lipid lowering,which might play their potential role by regulating PI3K-Akt signaling pathway,TNF signaling pathway and other metabolic pathways to play their role in lipid-lowering.(3)Biological effects of characteristic components on chronic colitisThe results of network pharmacology and molecular docking showed that AKT1,EGFR,SRC,MET,PIK3 CA,CASP3,TLR4,PTGS2 and other targets were the key targets of chronic colitis caused by characteristic components,which may play a potential role in regulating PI3K-Akt signaling pathway,HIF-1 signaling pathway and other metabolic pathways.Conclusion:In this paper,the chemical fingerprint method of Ligustri Lucidi Fructus was established,and the main fingerprint peaks were qualitatively analyzed.By comparing the fingerprints of the processing of Ligustri Lucidi Fructus with wine,26 characteristic components were selected,and the structural formulas of 7 of them were successfully identified.The difference analysis showed that 18 characteristic components after processing of Ligustri Lucidi Fructus with wine included m/z 288.1597,m/z 365.1192.The contents of m/z 430.1448,m/z 326.0630 and other eight characteristic components increased,while the contents of m/z 430.1448,m / z 326.0630 and other eight characteristic components decreased,and m / z 274.1442 and m / z 537.1780 were the two new components;From the perspective of network pharmacology and molecular docking methods,the mechanism of liver protection,lipid-lowering and chronic colitis induced by characteristic components was elaborated.m/z 274.1442、m/z365.1192 and other characteristic components through PI3K-Akt signal pathway,Erb B Signal pathway,HIF-1 signal pathway to protect liver;Through PI3K-Akt signaling pathway,TNF signaling pathway,c AMP and other pathways to reduce blood fat;m/z 430.1448,m/z 326.0630 through PI3K-Akt signaling pathway,HIF-1 signaling pathway,thyroid hormone and other signaling pathways to cause chronic colitis.To a certain extent,it explained the mechanism of enhancing efficiency and reducing toxicity in processing of Ligustri Lucidi Fructus with wine.
