Study Of Chinese Herb Extracts Interfere Helicobacter Pylori Vacuolating Cytotoxin Secretion

Helicobacter pylori is a gram-negative,spiral-shaped and microaerophilic bacterial pathogen which colony at the human stomach . Helicobacter pylori is associated with chronic gastritis and increased risk of gastroduodenal ulceration, gastric adenocarcinoma and mucosa-asssociated-lymphoid-tissue lymphoms. Several factors have been shown to be essential to bacterial colonization of the stomach mucosa. VacA is considered to be an important virulence factor in the pathogenesis peptic ulcer disease.VacA secreted by Helicobacter pylori acts inside cells and induces the formation of vacuoles and apoptosis,interacts with various components of the cell cytoskeleton,causing acting rearrangements and reduction of stress fibers.Our country posses rich Chinese medical resourse which have many advantages such as lower side-effect﹑drug-resistance and cure fee.So Chinese medicine has been one of the major medicine which can cure Helicobacter pylori infection,but the research of its acting mechanism and pathogenicity still remain vacant.Our research hypothsis that Chinese medicine such as Rhizoma Coptidis﹑Baical Skullcap Root can inhibit secretion of Helicobacter pylori VacA except its inhibition of biosynthesis and cell production,hence it can turn pathogenic Helicobacter pylori to non- pathogenic bacterium.Objective: To examine a possible influence of the extract of Chinese herb on the Helicobacter pylori vacuolating cytotoxin secretion in vitro.Methods: 1.Use the method of microscope observation and MTT to determinate the cytotoxic influence on HeLa cell by the extract of Chinese Herb .2. Determine the minimal inhibitory concentration(MIC)of the every kind of Chinese medicinal materials of Helicobacter pylori NCTC11638,and then take the sub-minimal inhibitory concentration of bacteria as the experimental concentration.3. The effection of Helicobacter pylori vacuolating cytotoxin secretion by the Extract of Chinese Herb: Determine the vacules of cells by microscope observation and stain cells by neutral red .4. Statistical methods: use SPSS11.5 to analyse the experiment data which expressed by mean±s ,t test .there have obvious difference when P<0.05.Results: 1. The effective medicine in vitro: Rhizoma Coptidis﹑Baical Skullcap Root﹑ Magnolia Bark﹑Areca Seed﹑Hawthorn Fruit﹑White Peony Alba﹑Corydalis Tuber.The vacuolation were decreased obviously after be effected by these medicine.Conclusion : 1. Rhizoma Coptidis﹑Baical Skullcap Root﹑Magnolia Bark﹑Areca Seed﹑Hawthorn Fruit﹑White Peony Alba﹑Corydalis Tuber can inhibit Helicobacter pylori vacuolating cytotoxin secretion effectively in vitro.
