Purpose:1. This paper discusses the clinical infection rate of H.pylori in Luoyang. 2. This paper explores the regnant genotype that infects H.pylori in Luoyang. 3. This paper analyzes the differences of regnant genotype that infects H.pylori in Luoyang among area, age and gender. 4. This paper evaluates the relationship between the genotypes that infect H.pylori and gastrointestinal diseases in Luoyang.Methodology: This paper collects 3495 samples of gastrointestinal patients who treated in the First Affiliated Hospital of Henan University of Science and Technology from September 2013 to April 2014 and examines the H.pylori using the 14 C urea breath test. Then this paper introduces the enzyme-linked immunosorbent adsorption test(ELISA) to detect Cytotoxin-associated gene A(Cag A) and vacuolating cytotoxin A(Vac A). In this step, the tested samples are 546 patients(male: 294, female: 252; age:10 to 79) who present positive results on 14 C urea breath test and the pathological diagnosis of gastroscope are chronic gastritis,peptic ulcers, duodenal bulb inflammation, the stomach, gastric polyps or reflux esophagitis. The next step is further test of Vac A subtypes for those who perform positive on Vac A test. The Vac A subtypes will be divided into Vac A-s1 a, Vac A-s1 b, Vac A-s2, Vac A-m1 and Vac A-m2 and finally using statistical analysis evaluates the data.Results:1. The H.pylori infection rate of 3495 gastrointestinal patients in Luoyang is 47.01%(1643/3495). Urban area makes up 41.66%(352/780) while county constitutes 48.54%(1318/2715). The infection rate in the urban area is lower than the county(P=0.04). When comparing H.pylori infection rate between two counties, the value of P always greater than 0.05 without significant difference.2. During the 546 cases of gastrointestinal disease patients which H.pylori positive in Luoyang, the positive rate of Cag A is 35.16%(192/546);the positive rate of Vac A is 48.35%(264/546), both are positive are 27.83%(152/546),melely positive of Cag A is 7.32%(40/546), melely positive of Vac A is 20.51%(112/546). In the 264 cases of Vac A positive gastrointestinal disease patients,the respective subtype Vac A positive rate is:sla:76.51%(202/264),slb:20.45%(54/264),s2:0.75%(2/264),ml:21.96%(58/264),m2:72.24 %(196/264), 6 cases undifferentiated type in s area, 10 cases undifferentiated type in m area, the sla:76.51% is highest in s area; the m2:72.24%is the highest in m area; the respective positive rate of Vac A gene subtype chimera are S1a/m1:15.9%(42/264),S1a/m2:50.00%(132/264),S1b/m1:3.03%(8/264),S1/bm2:13.63%(36/264),the dominate is Sla/m2 50.00%(132/264),the s2 chimera is unentdeckted.3. Comparing various regions in Luoyang, there is no significant difference in positive rate of Cag A, positive rate of Vac A, positive rate of Vac A subtype(sla, slb, s2, ml, m2) and positive rate of Vac A gene subtype chimera both between urban areas and counties and between two different counties. Moreover, the values of P are all greater than 0.05. Comparing various age groups, there is no significant difference in positive rate of Cag A, positive rate of Vac A, positive rate of Vac A subtype(sla, slb, s2, ml, m2) and positive rate of Vac A gene subtype chimera and the values of P are all greater than 0.05. Comparing different gender, there is also no significant difference in positive rate of Cag A, positive rate of Vac A, positive rate of Vac A subtype(sla, slb, s2, ml, m2) and positive rate of Vac A gene subtype chimera and the values of P are all greater than 0.05.4. Comparing different gastrointestinal diseases in Luoyang, there is no significant difference in positive rate of Cag A, positive rate of Vac A, positive rate of Vac A subtype(sla, slb, s2, ml, m2) and positive rate of Vac A gene subtype chimera.Conclusions:1. The rate of clinical H.pylori infection is 47.01% in Luoyang. The infection rate in the urban area is lower than the county, while there is no significant difference among counties.2. For the gastrointestinal patients who infected H.pylori in Luoyang, 48.35% of the Vac A test results are positive, 35.16% of the Cag A test results are positive and 27.83% of the patients are both positive on the Vac A and Cag A test. s1 a is the highest in s area, m2 is the highest in m area, s1a/m2 is the highest in Vac A chimeric. It can be inferred that the H.pylori infected by gastrointestinal patients in Luoyang take positive on Vac A sla/m2 as main genotype and most of them exist in the form of positive on Vac A and Cag A and Vac A sla/m2 chimera.3. There is no significant difference among the various regions, age groups and gender in terms of the positive rate of Cag A, Vac A and Vac A respectively, according to the infected H.pylori samples in Luoyang.4. The infected H.pylori samples in Luoyang have no significant difference among gastrointestinal diseases in terms of the positive rate of Cag A, Vac A and Vac A respectively. 5. The majority of infected H.pylori genotypes are weakly pathogenic in Luoyang.
