Objective: To investigate the effect of Ganyanping on the expression of hepatocyte growth factor (HGF) and transforming growth factor-β1 (TGF-β1) in rabbit with shchistosomal liver fibfosis and discuss the mechanism of Gyanyanping in further, so provide the theoretical basis for its clinical uses.Methods: The rabbit were separated randomly into three groups: normal group (control), model group (schistosomal infection + praziquantel) and Ganyanping group (schistosomal infection + praziquantel + Ganyanping). The rabbits of normal group were fed normally. The model group and the Ganyanping group were infected with cercariae of schistosomiasis japonicum (180 cercarie per rabbit) and treated with praziquantel (200mg/d) for two days at the end of the 6th week. And then Ganyanping group were treated with Ganyanping (1 tablet per day) for 6 weeks after the model was made successfully. The pathological changes of livers were observed by HE staining and electron microscope. The serum level of hyaluronic acid (HA),collagenⅣ(C-Ⅳ),laminin (LN) and precollagenⅢ(PCⅢ) were evaluated by radioimmunoassay (RIA). The expression of HGF and TGF-β1 were evaluated by immunohistochemical staining, Western blotting and RT-PCR.Results: Observed through HE staining, the schistosomal insect egg and fiber connective tissue in Ganyanping group were less than that of model group. The swelling of the liver cells of Ganyanping group was less than that of model group by electron microscope, and the deposition of fiber in Diss gap decreased. The indicators of liver fibrosis, such as HV,C-Ⅳ,LN and PCⅢin Ganyanping group, were lower than that in model group. Compared with the group M, the espression of HGF incressed,while the expression of TGF-β1 decressed in the liver in the group E. The difference was significant(P< 0.05).Conclusion: Our reaseach showed that Ganyanping could decrease the deposition of fiber and the expression of the indicators of liver fibrosis. Also Ganyanping could inhibit the expression of TGF-β1 and enhanc the expression of HGF in the liver of rabbit with schistosomal liver fibrosis, which may be one of the antifibrotic mechanism of Ganyanping.
