| Objective: To explore the expression and significance of TGF-β1,b FGF and HGF in hepatolithiasis-related intrahepatic cholangiocarcinoma.Methods:1.We choosed twenty-six cases of Hepatolithiasis-related Intrahepatic Cholangiocarcinoma,and twenty-eight cases of nonHepatolithiasis-related Intrahepatic Cholangiocarcinoma,and fifteen cases of Calculus of Intrahepatic Duct,and fifteen cases of normal tissue adjacent to Hepatic Hemangioma,who underwent hepatectomy or biopsy in the Department of Hepatobiliary and Pancreatic Surgery from January2012 to December 2020 and were finally confirmed by pathological biopsy.Immunohistochemistry(En Vision method)was used to determine the expression of TGF-β1,b FGF and HGF in tissue samples of each group.2.We choosed Masson’s three-color staining method to determined the content of collagen fibers in each group,and the degree of liver fibrosis was evaluated.3.We performed spearman analysis on the mean optical density of TGF-β1,b FGF,HGF and collagen volume fraction in the cholangiocarcinoma group to evaluate whether there is a correlation.4.We performed the mean optical density value and collagen volume fraction of TGF-β1,b FGF,HGF in the cholangiocarcinoma group and the clinicopathological characteristics of patients with cholangiocarcinoma were tested by independent samples T test to evaluate whether there are differences.5.In the cholangiocarcinoma group,We chooesd the mean optical density value of TGF-β1,b FGF,HGF and the collagen volume fraction and blood biochemistry were analyzed by spearman to evaluate whether there is a correlation.Results: 1.The expression levels of TGF-β1 in the four groups are in the order of HICC group,stone group,NHICC group,and normal group.The mean optical densities of the four groups are 0.063±0.034,0.029±0.009,0.019±0.007,0.002±0.003,the differences were statistically significant by one-way analysis of variance(P<0.0001).The expression levels of b FGF in the four groups were HICC group,HICC group,stone group,and the normal group.The mean optical densities of the four groups were: 0.051±0.021,0.05±0.022,0.027±0.009,0.02±0.008,The difference between the four groups by one-way analysis of variance,there was a statistical difference between HICC group and stone group,and the normal group(P<0.001),and the difference between the NHICC group and the stone group and the normal group was statistically significant(P<0.001)),there was no significant difference between HICC group and NHICC group(P>0.05).The expression of HGF in the tissues is in the order of stone and HICC group,stone group,NHICC group,and normal group.The mean optical density of the four groups is 0.084±0.021,0.074±0.017,0.059±0.022,0.02±0.005,there was a statistical difference between HICC group and NHICC group by one-way analysis of variance(P<0.001).2.The expression levels of collagen volume scores in the four groups were HICC group,NHICC group,stone group,and normal group.The collagen volume scores were 91.49±4.88,70.2±11.89,44.99±11.84,18.27±9.53,the difference between groups was statistically significant by using one-way analysis of variance(P<0.0001).3.The mean optical density of TGF-β1 in the cholangiocarcinoma group was positively correlated with the collagen volume fraction(P<0.0001).4.Collagen volume fraction is different from lymphatic metastasis and TNM staging of patients with cholangiocarcinoma(P<0.05).5.Albumin was negatively correlated with TGF-β1,b FGF,HGF expression and CVF(P<0.05);aspartate aminotransferase was positively correlated with HGF expression(P<0.05);prothrombin time was positively correlated with HGF and TGF-β1expression(P<0.05);Fibrinogen was positively correlated with HGF and TGF-β1 expression(P<0.05);lymphocytes were negatively correlated with TGF-β1 expression(P<0.05);CEA was positively correlated with HGF,TGF-β1 expression and CVF value Correlation(P<0.05);CA199 is positively correlated with b FGF,HGF expression,and CVF value(P<0.05);CA125 is positively correlated with b FGF,TGF-β1 expression,and CVF value(P<0.05);triglycerides are positively correlated with HGF,TGF-β1expression and CVF value were negatively correlated(P<0.05).Conclusion:(1)The expression of TGF-β1,HGF and liver fibrosis in HICC tissue is higher than that in patients with NHICC and intrahepatic bile duct stones.The expression of b FGF in HICC is not significantly different than that in NHICC,but it is significantly higher than that in intrahepatic bile duct stones.(2)The degree of fibrosis in cholangiocarcinoma tissue is positively correlated with the expression of TGF-β1.(3)The collagen volume fraction in cholangiocarcinoma tissue is positively correlated with lymphatic metastasis,and positively correlated with TNM staging,suggesting that liver fibrosis may be related to the prognosis of cholangiocarcinoma.In cholangiocarcinoma,TGF-β1,HGF expression and collagen volume fraction are positively correlated with the presence of combined stones,that is,there is liver fibrosis overexpression in stone-related intrahepatic cholangiocarcinoma. |
