| Paris polyphylla Smith(PPS), a traditional heat-clearing and detoxicating Chinese medicine, has the effects of antitumour, antidiabetic, anti-inflammatory and the inhibitory activity against platelet aggregation. Based on our traditional experience and modern research, the extracting and purifying process of total saponins from dried rhizome of PPS and the preparing process of the gel have been investigated.A content determination method has been established to mensurate the contents of total saponins, Polyphyllin I and Polyphyllin II.Using the yield of total saponins as the evaluation marker, single-factor experiments and orthogonal designs were performed to optimize the extracting process. The yield and the extraction rate of total saponins could be up to 4.24% and 94.01%, respectively. The total extraction rate of Polyphyllin I and Polyphyllin II was93.28%.The optimum purifying conditions were obtained by a combination of controlling the pH in alcohol solutions, alcohol extracting-water precipitating and macroporous resin techniques. Single-factor experiments and orthogonal designs were also conducted and the content and diversion ratio of total saponins were taken as the evaluation markers. The content and diversion ratio of total saponins in the effective fractions prepared by this optimum process were 71.6% and 85.71%, respectively.Membrane diffusion and transcutaneous penetration in vitro were used to determine the optimum base and penetration enhancers. |
