The experiment studied the effect of captopril used orally long-term on learning and memory and it inchuded three parts:1.We studied the effect of captopril used orally long-term on learning and memory in young mice. After given captopril one and a half months, the young mice were tested by water maze, avoid dark, step-down, then we killed the mice and did pathological test on their brains.The results showed that: compared with normal control group,after giving young mice captopril (7.59~8.79 mg / kg / day) for a month and a half,there wasn't significant changes in the index of water maze,such as latency,swimming distance,starting angle,,average velocity,times cross the platform in two minutes,total time in quadrant of the platform,total time in platform,distance in platform/total swimming distance percent,average velocity and the strategy of searching platform.In avoiding-dark test,the wrong times and latency of captopril group were not changed obviously on the 2th day. In step-down test, ,the wrong times and latency of captopril group were not changed obviously on the 1th and 2th day. The pathological examination showed that there were not obvious change in the cortex and hippocampus of captopril group compared with normal control group.2.The effect of captopril used long-term on learning and memory in normal adult mice were studied. Given captopril three months, the normal adult mice were examined water waze,avoiding dark and step-down ,then we tested cholinergic system (Ach content, AchE activity), free radicals and the activity of related enzymes (MDA, SOD, Na +-K + - ATP and Ca2 +-Mg2 +-ATP enzyme, GSH-Px) .The results showed that: administrated captopril(11.08~14.92mg / kg/day) three months later, the index of captopril goup mice were not change significantly on water maze,avoiding dark and step-down compared with normal control . The biochemical results showed that content of protein and Ach in the brain were significantly increased (P <0.01), the content of AchE(P <0.05) was manifestly step up and the content of MDA cut down (P <0.05), SOD, GSH-PX did not change obviously.The trendency of activity on Ca2 +-Mg2 +-ATP enzyme and Na +-K +-ATP enzyme declined, but there were not obvious change in statistical methods. We concluded that it would step up the function of learning and memory in normal adult after taking Captopril for a long time.3.Captopril used long-term have effected on learning and memory in vascular dementia rats. We made vascular dementia model by permanent bilateral carotid artery ligation,and observed the effect of captopril on learning and memory of vascular dementia rats in behavior and pathology.We examinated the cholinergic system of brain ( Ach content, AchE activity), free radicals and the activity of related enzymes (MDA, SOD, Na +-K +-ATP and Ca2 +-Mg2 +-ATP enzyme, GSH-Px).The results showed that: After given captopril(7.52~10.48mg / kg / day) two months, captopril group in water maze,avoiding dark and pathology have no significant change on vascular dementia rats. Biochemical index indicated that compared with model group, the activity of AchE in the brain increased (P <0.01), brain protein,Ach,MDA,SOD,GSH-PX,Na +-K +-ATP and Ca2 +-Mg2 +-ATP enzyme didn't have significant change. We concluded that it didn't affect the function of learning and memory on vascular dementia rats obviously after used Captopril long-term.
