Lung cancer is mostly reason which lead cancer patients death at present.Some experts announced that lung cancer and aids will be the most harmful disease related to bad habits to human health in this century.In our country lung cancer will be notable increase in a long time.Since the new diagnosis treat methods and anticancer drug emergence,human acquired tremendous progress in anticancer, but in some large city cancer has the highest mortality rate among all the cancers . To study cancer at the Molecular level has been hotspots in these years.Some discoverys of cancer connected genes made early diagnosis and treat of cancer possible.Some study found that Novel genetic Polymorphism in anti–oncogene is important reason lead cancer,so theO~6- methylgu- anine-DNA methyltransferase(MGMT) which is the only enzyme repair Polymorphi- sm in human body has been hotspots these years.SNP(single nucleotide polymorphism),which is the third generation genetic marker showed to have the characteristics of high-density inheritance stability and facilitation in analysis automation.give ours a handy way to study MGMT.Our investigation means to evaluate the association between MGMT SNP and susceptibiliy in NSCLC(non-small cell lung cancer)by case -control test.Blood samples were collected from 48 patients with histological proven diagnosis of NSCLC recruited at the department of thoracic surgery,No.2 clinical hospital of Jilin University.Blood sample of 50 healthy donors were collected as control.Then the genome DNA was extract from blood sample and PCR(polymerase chain reaction)amplification of the fragment involving the MGMT codon 53 RFLP was performed and the PCR product is 235bp,the PCR product of Following the PCR the PCR product were digested with Hpy188I . The result of our investigation were analysed statistically.We found that C/C genotype frequency was higher in NSCLC patients than that of the healthy controls(χ~2=0.36,P>0.05).T/T genotype frequency was lower in NSCLC patients(χ~2=0.002,P>0.05).C/T genotype frequency was lower in NSCLC patients(χ~2=0.21,P>0.05). The Conclusion:In the Northern Chinese population MGMT codon 53 SNP has no associated with the susceptibility of NSCLC.
