| Flemingia macrophylla is the root of Leguminous plants of Flemingia Roxb . ex Benth.The extraction,separation and identification of the active components from the Flemingia Philippinensis were studied first.And then,their activities in bacteriostasis and thrombosis prevention and treatment were also done.First,we extracted Flemingia Philippinensis with water and 70% ethanol,got water extract which weighted 120g and ethanol extract which weighted 150g. Water extract was then degreased by aether,abstracting with ethyl acetate was following,at last, ethyl acetate extract which weighted 45g was got. One compound was isolated and identified by TLC and HPLC,we recognized it as chlorogenic acid,which was first separated from Flemingia Philippinensis. Ethanol extract was isolated by amberlyst, which was gradient eluted by the ethanol.We collected the 70% ethanol extraction,which weighted 38.4g,as the flavones of Flemingia Philippinensis(FFP).Also one compound was isolated and identified by TLC and HPLC,we recognized it as genistein.Used the genistein as the reference standard ,we determined the concentration of FFP,which was 48.9%.Second, the antibacterial activities of the extraction from Flemingia Philippinensis was studied,the strains that be tested were Escherichia coli, Staphylococcus aureus, Aeruginosus Bacillus and Blastomyces albicans. The minimal inhibitory concentration(MIC) of the water extract (FWE) and the alcohol extracts (FAE)against Escherichia coli was 0.5g/ml;The MIC of the FWE and the FAE against aeruginosus Bacillus was 0.25g/ml; the FWE had inhibitory action against Staphylococcus aureus in the range of concentration(0.03~0.5 g/mL),however,it had no inhibitory action against Blastomyces albicans in the range of concentration. The MIC of the FAE against Staphylococcus aureus was 0.06g/ml.It also had inhibitory action against Blastomyces albicans.The MIC was 0.5g/ml.The activities of thrombosis prevention and treatment in rats by FFP were studied at last. The indexes that be observated were carotid thrombosis in rats which were stimulated by electronic;bleeding time in tails tip of rats and haemorheology indexes in rats with femoral artery thrombo. The results showed that FFP can prolong the occlusion time of the carotid thrombosis in rats that stimulated by electronic; the bleeding time in tails stip was prolonged,too;,and haemorheology indexes in rats with femoral artery thrombo also had changing obviously.The result of this theses supplied the application value of Flemingia Philippinensis in treating the gynecologic sickness and prevention and treatment of the thrombus.And we also lied a foundation for the development and utilization of Flemingia Philippinensis. |
