ObjectiveBy the research of BRCA1 and FANCD2 in breast cancer's expression, detect the expression of BRCA1 in breast cancer to find if the expression of BRCA1 in familial breast cancer is higher than sporadic ones and to detect the expression of FANCD2 in breast cancer to find if the expression of FANCD2 in sporadic breast cancer is higher than familial ones. Analyze the expression of BRCA1 and FANCD2 in family and sporadic breast cancer, we want to know the relations between the BRCA1 and FANCD2 genes and if familiar breast cancer makes its effect through the BRCA1/FANCD2 pathway,in order to make sure the sense of the BRCA1 and FANCD2 expression, the familial breast cancer in particular.MethodsAll breast cancer surgery specimens come from the tumor surgery department of the China Medical University from January 2001 to August 2007. the total cases is 60, including 30 cases of familial breast cancer and 30 patients of sporadic breast cancer. The patients who have breast cancer first degree relatives are belong to the familial breast cancer group. The patients who have no breast cancer relatives are belong to the familial breast cancer group. All the patients were female patients, who have pathology of invasive ductal carcinoma, and without preoperative radiotherapy and chemotherapy. The age span from 41 to 65-year-old. Mouse anti-human monoclonal antibody BRCA1, SP kit and DAB were purchased from Fuzhou Maixin biotechnology development company. FANCD2 antibody was purchased from SANTA CRUZ biotechnology development company. The experiment was completed in the Gastric Cancer Research Center of the First Affiliated Hospital of China Medical University by immunohistochemical SP method. After all wax block sectioned, the 60℃drying, dewaxing, hydration, hydrogen peroxide closed endogenous peroxidase, high temperature and high pressure processing, antigen retrieval, cooling, adding peroxidase inhibitor A serum ,the first antibody B,the second antibody C, D reagents, the final DAB color, hematoxylin-stained, dehydration cementing, optical microscopy. Analyze the expression of BRCA1 and FANCD2 in family and sporadic breast cancer, use SPSS software to analyze the results.Results1,Positive rate of BRCA1 protein expression in familial breast cancer is 70.0%(21/30), in sporadic breast cancer is 13.3%(4/30). There was a difference between the groups (P < 0. 05). 2,Positive rate of FANCD2 protein expression in familial breast cancer is 40.0%(12/30), in sporadic breast cancer is 66.7%(20/30). There was a difference between the groups (P < 0. 05) .3,In familial breast cancer, positive rate of FANCD2 expression in those positive expressions of BRCA1 protein is 36.7% (11/30), in negative expressions of BRCA1 protein is 3.3% (1/30). (P < 0. 05) .4,In sporadic breast cancer, positive rate of FANCD2 expression in those positive expressions of BRCA1 protein is 13.3% (4/30), in negative expressions of BRCA1 protein is 53.3% (16/30) . (P > 0. 05).Conclusion1,Expression of BRCA1 in familial breast cancer is higher than that in sporadic breast cancer. It means BRCA1 playing a very important role in the familial breast cancer.2,Expression of FANCD2 in familial breast cancer is lower than that in sporadic breast cancer.This indicate that the FANCD2 gene play a role in the course of forming of breast cancer as an anti-oncogene. 3,Expression of FANCD2 has a negative relationship between the familial breast cancer and the sporadic ones. This demonstrate that the Fanconi anemia / BRCA pathway play a important role in breast cancer and especially the familiar breast cancer.4,According to the expression of BRCA1 and FANCD2,we can evaluate the affectability and risk of familial breast cancer.
