The Effect Of Luoshi Neiyi Prescription On Apoptosis And Matrix Metalloproteinase Of Uterine Ectopic Endometrium

Objective:To observe the influence of Luoshi Neiyi Prescription on MMP-9,MMP-14 and Bax expression levels of uterine ectopic and normal endometrial cells,according to which study the therapeutic mechanism on endometriosis in apoptosis and extracellular matrix degradation,and provide a more effective and practical method and scientific basis on it.Research Methods:1.Study Group(EE):ectopic endometrium of 15 patients with endometriosis; Control Group(NE):10 cases of normal endometrium.2.Culture of single cells which were degested from endometria tissus with 0.25% trypsin,0.1%collagenase,and then added pharmacological serum,blank serum into Medium respectively,collected the cells after 24h.3.Total RNA was extract by using TRIZOL;The different expression of MMP-9mRNA,MMP-14mRNA,baxmRNA was then observed through reverse transcriptase polymerase chain reaction(RT-PCR).Results:1.Cell culture showed that 12 cases of normal endometrium and 8 cases of ectopic endometrium were successfully cultured;they exhibited similar appearances and the same cytoskeleton system under inverted microscope.2.The expression of MMP-9mRNA,MMP-14mRNA,baxmRNA: 2.1 Ectopic endometrium expressed higher Bax(P＜0.05),compared with normal endometrium,;the expression of MMP-9,MMP-14 were just the opposite(P＜0.01). 2.2 In normal endometrium at proliferative phase or secretory phase,Bax,MMP-9 and MMP-14 were all expressed.There were no significant difference in expression among the three groups(P＞0.05); 2.3 In ectopic endometrium,the herbs in drug serum group vary significantly higher expression of Bax(P＜0.01),lower expression of MMP-9 and MMP-14 (P＜0.01),Compared with non-medicated serum and blank control group. 2.4 In ectopic endometrium,the expression of Bax and MMP-9,MMP-14 had no obvious correlation(P＞0.05).Conclusion:1.The higher expression of MMP-gmRNA,MMP-14mRNA in the ectopic endometrium ofwomen with endometriosis,facilitate the ectopic growth of the endometrium, precipitate the process of the endometrioses;while the BaxmRNA is in low expression,which prevents apoptosis of the ectopic endometrium and keeps it grow.2.Bax and MMP-9,MMP-14 have a close relation with the occurrence of endometriosis,yet there is no significant relationship in their expression level.3.Luoshi Neiyi Prescription can induce apoptosis of the ectopic endometrium, make it spontaneous regression,and do not damage the normal tissue as a result to reach the therapeutic aim.In addition,the mechanism of Luoshi Neiyi Prescription may have relationship with depressing the expression of MMPs and reducing the degradation of the extracellular matrix.
