| ObjectiveSet up rat models with endometriosis (EMs) by adopting autologous grafting and study the effects on the endometrial receptivity of the experimental rats delivered by EMS. Study the differences of index of rat’s in ectopic pinocytosis tu, integrin av beta3, the expression of female, progestational hormone in serum after applying Luoshi Neiyi Prescription to the rat models and explore mechanics concerning how Luoshi Neiyi Prescription affect endometrial receptivity of the rats with EMS and the effect’s target spot. Apply the model to clinical treatment and record the effects delivered by Luoshi Neiyi Prescription on II EMs infertile patients concerning the endometrial receptivity, with the purpose of providing a new basis for the above-mentioned treatment.MethodsExperiment:Purchase40SPF mature female SD rats, which were randomly divided into3groups, respectively, the group treated with Luoshi Neiyi Prescription, the model group and the blank group. The blank group, unlike other two groups, would not set up EMS rat models by adopting autologous grafting. After building the models, Luoshi Neiyi Prescription would be applied to the group treated with Luoshi Neiyi Prescription, the other with sterilization water treatment for8weeks. On the fourth pregnant day after stopping drugs, the right side of the uterus of rats were taken to observed with electronic microscope concerning the expressions of lining pinocytosis tu, SABC method to detect lining integrin av beta3, ria method to detect the expression of female progestational hormone in serum.Clinical part:choose30patients confirmed to be II EMs infertile patients by taking laparoscopy and randomly divide them into two groups, respectively, the roche group and the control group. Dose patients with drugs for3consecutive cycles and observe the improvement of clinical symptoms of each group, and monitor each postoperative follicular development in dynamic state and render guidance of fertilization as well as takeing pregnancy rates of each groups during treatment for statistical analyze. Upon completion of treatment, on the8days following with ovulation cycle, take a little lining of not pregnant patient and observe its integrin av beta3expression and monitor the female progesterone levels in serum. ResultsExperiment:1.The rate of successfully building up rat models with endometriosis (EMs) can reach92.59%by adopting autologous grafting..2.After comparing the rats’in ectopic integrin av beta3COD value, P>0.05, which was of no statistics significance.3. As for the rats’mature pinocytosis tu expression, it can be concluded that P<0.05when comparing firstly between the Luo Shi Nei Yi group and model groups, and secondly between the model groups and blank groups, which was of statistics significance. However, when comparing the Luo Shi Nei Yi group and the blank groups, P>0.05, which was of no statistics significance.4. It can be observed that the development of rats’ mature pinocytosis are consistent concerning roche groups and blank groups when comparing the morphology characters of rats’mature pinocytosis during implantation window period, which are characterized by the following features:pinocytosis is rich, of uniform distribution and with clear boundaries. The mature pinocytosis dash forward of the model group rats is of small bumps and uneven distribution and the size differs from each other.5. When comparing the estrogen expression level between two groups, P>0.05, according to the alpha=0.05, the difference is not significant. P>0.05, differences between each group had no statistical significance.6. it can be found that P<0.05when comparing progesterone expression level between three groups, with statistically significant. The difference is of statistically significant when comparing between roche group and model group, which can be concluded that P<0.01, and between the blank group and model group, which can be found that P<0.05. However, there was no statistically significant difference when comparing the roche group and the blank group, which can be found that P>0.05.7. development situation of the pinocytosis and serum progesterone level have significant correlation (r=0.646, P<0.01), which were positively correlated; Development of pinocytosis and serum estrogen had no correlation (r=0.173, P=0.320). Development of Pinocytosis is negatively correlated with the female serum progesterone ratio (r=0.336, P=0.336).Clinical part:1.The clinical symptom score of roche group had improved before and after receiving treatment, and there was statistically difference when comparing with the control group. Meanwhile, the pregnancy rate of two groups has no statistical significance. Two groups of patients with pregnancy were intrauterine pregnancy.2. When comparing the two groups concerning estrogen, P>0.05and there was no statistical significance, indicating that there was no difference between two groups on the expression of the estrogen level. Progesterone comparison was statistically significant (P<0.05), suggesting the two groups’ progesterone level expression were of statistically significance.3. The comparison between two groups of endometrium integrin av beta3COD value demonstrated that P<0.05, with statistical significance. Integrin av beta3COD value of the patients of roche group is higher than that of the control group.ConclusionExperiment:1.The rate to build up a successful rats models with Ems is high by adopting the methods of autologous grafting and filling estradiol valerate, The methods are operative and repeatable, which are suitable for experiment and study requiring large quantities of samples.2. EMs can affect the rats’pinopodes through a certain way, lowering the endometrial receptivity of Ems rats. The Luo Shi Nei Yi Prescription can improve the endometrial receptivity of Ems rats by increasing serum progesterone level.3. Pinopode is of positive correlation with serum progesterone level. No relationship is observed between the development of pinocytosis and serum estrogen levels. The development of pinocytosis is associated with the ratio of E2/.P, and the high levels of E2/P inhibits the development of pinocytosis. The best ratio remains to be further study.Clinical part:1. Symptoms of Stage Ⅱ endometriosis infertile patients improved significantly after receiving the treatment of Luo Shi Nei Yi Prescription. The pregnant rate was29.42%during the treatment.2. During the period of ovulation cycle, the in ectopic integrin av beta expression of Ⅱ vision disorder infertile patients is improved and progesterone also rises accordingly, The specific mechanism and targets await further research and study. |
