Research Of Anti-Drug Dependence Effect And Mechanism Of FZY And SDM

Objective:To research the therapeutical effect and its mechanism of FZY and SDM on morphine-dependent macaca rhesus.Method:90 days of morphine-dependent rhesus monkey model was induced by administration(s.c.)of morphine at a gradual increasing dosage.The withdrawal symptoms,changes of weight and body temperature were recorded. Serum specimens of rhesus monkey were collected before the experiment begins, after the model established and after the treatment with drug.Change of concentration of nitric oxide before and after withdraw was tested by nitrate reductase.Change of activity of nitric oxide synthetase before and after withdraw was tested by kit.Change of concentration of dopamine before and after withdraw was tested by high performance liquid chromatogramfluorescence detection.Result:①Withdrawal symptoms could be inhibited significantly by FZY at the dosage of 1.644 and 3.288 g·kg^-1(P＜0.05).Weight lose could be controlled from third day of withdrawal by FZY at the dosage of 1.644 and 3.288 g·kg^-1(P＜0.05).Concentration of nitric oxide and dopamine and activity of nitric oxide synthetase could be reduced by FZY at the dosage of 1.644 and 3.288 g·kg^-1(P＜0.05 and P＜0.01).②Withdrawal symptoms could be inhibited significantly by SDM at the dosage of 2.4 and 4.8 g·kg^-1(P＜0.05). Concentration of nitric oxide could be reduced by SDM at the dosage of 4.8 g·kg^-1(P＜0.05).Activity of nitric oxide synthetase could be reduced by SDM at the dosage of 2.4 and 4.8 g·kg^-1(P＜0.05 and P＜0.01).Conclusion:FZY and SDM in suitable dosage has therapeutical effect on morphine-dependent rhesus monkey by alleviating withdrawal symptoms and controlling the weight loss with the possible mechanism of down regulation the level of NO by inhibiting NOS activity and reducing the concentration of dopamine in vivo.
