The Protective Effect Of Keratinocyte Growth Factor On Mice Irradiated By ⁶⁰Co Gamma Ray And The Primary Research About Its Mechanism

Objective:To investigate the anti-irradiation effect of KGF on mice irradiated by 60Co gamma ray and then make some primary research of its mechanism on the basis of the results in this experiment.Methods: Sixty Kumming mice were randomly divided into six groups, including 60Co model group, KGF groups (low dose, middle dose, and high dose) and control group, Geniposide positive control group. Mice were irradiated with 6 Gy 60Co gamma rays(1Gy/min)after administration of KGF and Geniposide for 7 days. The weight of each group was calculated at the 1st, 7th and 12th day of the experiment; Six days after the irradiation, the spleen and thymus indixes were calculated; the section of liver, lung and jejunum were observed; the amount of superoxide dismutase (SOD), glutathione peroxidase (GSH-px), glutathione S-transferase (GST), malondialdehyde (MDA), and total antioxidative capacity (T-AOC), as well as the length of jejunum intestinal villus were detected.Results:1) KGF could amelirotate the discrease of diery and weight of mice induced by the irradiation of 60Co gama rays;2) Compared with control groups, KGF could obviously increase the spleen and thymus indices, ameliorate the radiation damage in spleen and thymus;3) KGF had a positive protective effect on the irradiation injury in liver, lung and jejunum of mice;4) KGF could inhibit the decrease of SOD, GSH-px, GST and T-AOC activities and the increase the amount of MDA dose-dependently in lung and liver;5) the length of the jejunum intestinal villus of the mice treated with KGF and Geniposide were obviously longer than that of the 60Co group.Conclusions: KGF could protect mice irradiated by 60Coγrays. The mechanisms might be related to its anti-oxidative capability and the modulation of the immune system.
