Cichoric acid,a powerful stimulant for the immune system,is widely used in Europe and America.Cichoric acid is extracted from natural plant usually from Echinacea Purpurea.At this article cichory is used as raw material,owing to its safety.It is quite popular for people to have cichory instead of vegetable in their dinner.Morover,cichory is in the list of raw material published by ministry of health of China,in which all materials are not only food but also medicine.It is significant to study the extraction of cichoric acid from cichory.To compare the chicoric acid contents of cichory,including Cichorium glandulosum Boiss Et Hout and C.intybus L,it has been found that there are differences with the contents of chicoric acid in various species,different part and even different producing area.The significant difference was obvious in C. glandulosum Boiss Et Hout and C.intybus L.by cluster analysis.The highest amounts of cichoric acid were measured from the part of up-ground of C. intybus L.which is usually discarded.So the results showed that the up-ground part of C.intybus L.was the optimum material for preparing chicoric acid.Solvent extraction and ultrasonic extraction were studied in extracting cichoric acid from cichory.The optimal conditions of solvent extraction obtained by equality design were as follows:at 90℃with 54%ethanol(24:1, v/w),for 1h.The optimal conditions of ultrasonic extraction were obtained by orthogonal design.If 10g raw materials floading in 120ml 54%ethanol were treated 30 minutes with 40W ultrasonic,we can gain maximum extraction. Compared with traditional technics,ultrasonic extraction technic was rapid,no heating and fewer solvent.In order to shorten treating time,samples can be treated by ultrasonic before mensuration.The purification process was investigated.When the crude chicory extraction purified one time with acetic ether(1:1,v/v)at 60℃,the highest purity of cichoric acid can be obtained.At above contidion,extraction ratio was over 84%.It has been found that there are some effect on cichoric acid content by using different style of drying.The effect of spraying drying and of freezing drying are similar,but better than that of vacuum drying. In addition,cichoric acid standard was obtained by high performance liquid chromatography.The highest purity of cidhoric acid was over 95%.The HPLC fingerprint of cichory extraction is established.There were 8 mutual peak in fingerprint which can be found in chromatography of the up-ground part of C.intybus L..In fingerprints of 10 bacthes cichory extraction,the coefficient of correlation were more than 0.997 and cosine value of vectorial angle were more than 0.998.High similarity showed that the process of cichory extration was stabilization.The average of cichofic acid content was 7.22%in ten bacth extraction,therefore it can be used as raw material of functional food.
