Study On The Effect Of Bone Marrow Mesenchymal Stem Cell And It's Lysate Transplant Juncture Hyperplasy Correlation Factor Which Promote Vascularization In Infarcted Myocardium

Objective: Study on the effect of bone marrow mesenchymal stem cell and it's lysate transplant juncture hyperplasy correlation factor which promote vascularization in Infarcted myocardium。Methods: MSCs from SD mice was acquired by density gradient centrifugation and adherence screening method. Cultured in vitro,to get the 3-thd passage MSCs,get the MSCs lysate;take the Peripheral blood to determine the density of VEGF,FGF-2,SDF-1 in different time point after MI;To find the peak time density of three cytokine secrete after MI. proceed the transplantation of MSCs,MSCs lysate,the mixture of MSCs and MSCs lysate,culture media after MI;The animal which suffered acute myocardial infarction be distinguished to four groups:control group,Stem Cell Transplantation group and Stem Cells lysate group,the mixture of MSCs and MSCs lysate group. sacrifice the animals 4 weeks after the transplantation, use rt-PCR and immunohistochemisty to check out the expression of VEGF in the infarction tissueS of MI.Results: In peripheral blood,VEGF express maxlmum in 7day after MI, bFGF express maxlmum in 3day and their densities fall-off after that time, but the SDF-1 have the maxlmum density in 24hour after MI and fall-off after that time. Transplant MSCs and It's lysate in different time point after MI,the ability of promote the expression of vascular endothelial growth factor in the MSCs group and the MSCs lysate group are better than the culture media group,but there is no difference between the MSCs group and the MSCs lysate group.The groups which Transplant the MSCs and the MSCs lysate 1 week after MI,is better than Transplant the MSCs and the MSCs lysate 1h,24h,3d after MI.Conclusion:VEGF,bFGF,SDF-1have visible density curve linearity after MI, possible have obviously dependablity to the recovery of infarctus myocard and Angiogenesis after MI; MSCs lysate also have the effect of hyperplasy correlation factor which promote vascularization in Infarcted myocardium and have no difference with MSCs;the mixture of MSCs and MSCs lysate has a better effect to promote vascularization in Infarcted myocardium than only MSCs and MSCs lysate;The groups which Transplant the MSCs and the MSCs lysate 1 week after MI,is better than Transplant the MSCs and the MSCs lysate 1h,24h,3d after MI to Promote the generate of blood vessel in infarced myocardium.
