Objective:To investigate the roles of Phosphatidylinositol 3-kinase(PI3K) in the regulation of expressions of Osteopontin(OPN)and vascular endothelial growth Factor(VEGF)in hepatocellular carcinoma(HCC) cells.Methods:The high metastasic cell line HCCLM3 were cultured in DMEM medium,LY294002 is the specific inhibitor of PI3K.The expressions of OPN mRNA and VEGF mRNA in HCC cells were detected by reverse transcriptional polymerase chain reaction(RT-PCR) after incubating with LY294002.Western Blot was used to research on the expressions of OPN in HCC cells.Results:1.In HCCLM3 cells,the expressions of OPN mRNA and VEGF mRNA can be apparently inhibited by LY294002.With the increase of LY294002 concentration,the expressions of OPN mRNA and VEGF mRNA decreased gradually(P<0.01)and in a dose- dependence manner.2.The expressions of OPN mRNA and VEGF mRNA can be apparently inhibited by LY294002.With the increase of time,the expressions of OPN mRNA and VEGF mRNA decreased gradually(P<0.01)and in a time-dependent manner.3.With the increase of LY294002 concentration and time,the expression of OPN protein decreased gradually(P<0.01)and in a dose-dependence and time-dependent manner.Conclusion:1.LY294002 can reduce the expressions of OPN and VEGF in HCCLM3 cell line,the expression of OPN and VEGF in HCC cells is regulated by PI3K.2.OPN maybe relate to the metastasis and recurrence of HCC.
