AIM:To investigate the effects of HDAC6 overexpression on the proliferation and apoptosis of human gastric cancer cell line SGC-7901 and to study whether HDAC6 can be used as a target for therapy of gastric cancer.METHODS:Human gastric cancer cell line SGC-7901 and BGC-823 was cultivated by routine method,then Determine HDAC6 expression gastric cancer cell lines by immunocytochemistry to screen a cell model for further studies.HDAC6 eukaryotic expression vectors pCDNA3.1-HDAC6-FLAG were transfected into SGC-7901 by lipofectamineTM2000,the transtected cells were screened by G418.Western blot was used to detect the HDAC6 protein expression level in the transtected cells.The proliferation of transtected cells was examinedby MTT assay and Colony formation experiment,and the cell apoptosis induced by cisplatin(CDDP)were detected by flow cytometry(FCM).RESULT:(1)The HDAC6 protein was found both in SGC7-901 and BGC-823 detected by immunocytochemistry,but the positive rates of HDAC6 expression in SGC-7901 were markedly higher than in BGC-823(0.28±0.07 vs 0.19±0.04,P<0.05).(2)Western blot revealed that HDAC6 protein expression levels in SGC-7901 cells transfected by pCDNA3.1-HDAC6-FLAG were remarkably increased in comparison with SGC7901 cells transfected by pcDNA3.1 empty vector and SGC-7901 cells, respectively(3591.52±237.65 vs 1899.93±133.43,1685.59±176.70,P<0.05).(3)Compared with the control group,the proliferation of the SGC-7091 cells transfected with pCDNA3.1-HDAC6-FLAG were enhanced signifcantly(P<0.05).(4)The apoptotic rates of the SGC-7091 cells transfected with pCDNA3.1-HDAC6-FLAG were decreased obviously[(10.46±0.65) %vs(20.59±0.88)%,(22.31±1.40)%,P<0.05].CONCLUSION:HDAC6 was stably expressed in the gastric cancer cell,its overexpressionmay enhance proliferation of gastric cancer cell,and decrease gastric cancer's sensitivity for regular chemotherapy agents such as cisplatin(CDDP).HDAC6 may be a potential target for therapy of gastric cancer.
