| Objective:To investigate the role of RhoA/Rho kinase system in the onset of labor.Methods:20 cases in labor(L group).20cases not in labor(NL group) but in term pregnancy, and 20 cases of non-pregnant(NP group)women undergoing hysterectomy due to cervical intraepithelial neoplasia were recruited into this clinical study.Real-time quantitative polymerase chain reaction and Western Blotting methods used to investigate the expression of RhoA and ROCKI mRNA and the corresponding protein.Results:1)The expression of RhoAmRNA in L group[(3.51±0.56)×10-4]was significantly higher than that in NL group[(2.81±0.52)×10-4],P<0.05,which was significantly higher than that in NP group[(2.11±0.54)×10-4],P<0.05.2)The expression of ROCKI mRNA in L group was[(4.07±0.66)×10-3]was significantly higher than that in NL group[(2.71±0.31)×10-3],P<0.05,which was significantly higher than that in NP[(2.01±0.23)×10-3],P<0.05.3)The expression of RhoA protein in the L group,NL group,NP group were(0.72±0.13),(0.64±0.17),(0.58±0.14).There was not significantly difference in among different groups,P>0.05.4)The expression of ROCKI protein in L group (0.56±0.14),was significantly higher than that in NL group(0.42±0.11), P<0.01,which was significantly higher than that in NP group(0.29± 0.08),P<0.01.Conclusion:The expression and activity of RhoA/Rho kinase in the uterine smooth muscle may play an important role in the onset of labor. |
