Lung cancer is a major health hazards for human, but there is no available marker for lung cancer identification. Previous studies showed it expressed at a high frequency in lung cancer, it's localization in cell is different at different stage in tumor. Objective Analysis the expression circumstance of Stk33 in different staging lung cancer and normal lung tissues. To study the expression dependablity abnormality of Stk33 in lung cancer. To study the expression rule of Stk33 and the significance about the development and prognosis in lung cancer.Methods We evaluated the quantitative expressions of Stk33 mRNA in 8 lung cancer cells and lung cancer tissues samples from patients who had undergone surgery between the October of 2008 and the June of 2009 using real-time quantitative reverse transcriptase polymerase chain reaction (real-time quantitative RT-PCR). At the same time, we evaluated the qualitative expressions of Stk33 protein in lung cancer tissue samples using immunohistochemistry (IHC) and Western-blot.Results1. The real-time quantitative RT-PCR study demonstrated that the expressions of Stk33 mRNA in cells of lung cancer were significantly correlated.2. The Western-blot study demonstrated that the expressions of Stk33 proteinum in cells of lung cancer were significantly correlated.2. The immunohistological study indicated that Stk33 were expressed as buffy granules not only in the cytoplasm of cancer cells, but also in macrophages in lung cancer. The positive rates of Stk33 protein in specimens of lung cancer were significantly higher than those of the peritumoral and pulmonary benign diseases tissues and the proteins aforesaid were correlated remarkably with lymph node metastasis, histology type, cell differentiation degree and PTNM stage of lung cancer. The immunohistological study of 124 lung tissues manifested that the positive rates of Stk33 protein in the positive lung cancer tissue were significantly higher than those in the negative normal lung tissue. According to the correlation analysis, the expressions Stk33 protein in lung cancer were correlated with lymph node metastasis. The Stk33 protin in the cancerous invasive edge of lung cancer were significantly higher than those in the center of cancerous tissues and those in the pulmonary benign diseases tissues. Correlation analysis demonstrated Stk33 protin is dependability with the lung cancer tissues exist and lymphaden aversion. The expression of Stk33 in the cancerous invasive edge of lung cancer were significantly higher than those pulmonary benign diseases tissues.
